摘 要:目的:研究中国汉族人群性激素结合球蛋白(Sex hormone-binding globulin,SHBG)基因Asp327Asn多态性与2型糖尿病(Type 2 diabetes mellitus,T2DM)发病风险的关系。方法:应用限制性片段长度多态性(Polymerase chain reaction-restriction fragment length polymorphism,PCR-RFLP)方法,对599例重庆及周边地区汉族人群(T2DM患者248例、正常对照351例)Asp327Asn进行基因分型。用稳态模型(Homeostatic model assessment,HOMA)评估胰岛素抵抗(HOMA-IR)和胰岛β细胞功能(HOMA-B)。结果:Asn等位基因的分布频率T2DM组低于正常对照组(分别为14.5%、20.1%);与Asp等位基因携带者相比,Asn等位基因携带者患T2DM的风险显著降低(OR=0.62,95%CI为0.42~0.91,P=0.016)。结论:SHBG基因Asp327Asn位点Asn等位基因可能与低T2DM患病风险相关。Objective:To investigate the association of a common functional polymorphism(Asp327Asn) in the sex hormone-binding globulin(SHBG)gene with type 2 diabetes mellitus(T2DM) in Chinese Han population.Methods:Asp327Asn polymorphism in 248 cases(T2DM group) and 351 controls(NC group) were genotyped by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) technique.Homeostatic model assessment(HOMA) was used to assess beta cell function(HOMA-B) and insulin resistance(HOMA-IR).Results:The variant Asn allele frequency was significantly lower in T2DM group than(14.5%,20.1%,respectively) that in NC group.Unconditional logistic regression analysis showed that the variant Asn allele was associated with a reduced risk of T2DM(OR=0.62,95%CI:0.42~0.91,P=0.016).Conclusion:This study suggests that the Asn allele in the SHBG gene might be related to a reduced risk of T2DM in the Chinese Han population.
