Effect of QiangxinChongji on Myocardial Apoptosi and Ventricular Remodeling in Rabbit Model of Heart Failure

摘  要:目的:建立家兔充血性心衰模型,以探讨强心冲剂在调节细胞凋亡及抑制心室重塑方面的作用。方法:将60只家兔随机分为6组,假手术组、模型组、地高辛组、低、中、高三个剂量强心冲剂组,每组lO只,采用腹主动脉缩窄术建立家兔CHF模型,予强心冲剂和地高辛灌胃9天,免疫组化方法测定心肌细胞凋亡相关基因表达的变化。结果:心衰家兔Bax表达明显增加,与此同时Bcl-2及肌动蛋白(α-actin)表达明显降低;而与模型组相比,强心冲剂组和地高辛组则明显下调Bax蛋白表达,上调Bcl-2及肌动蛋白表达,并且强心冲剂作用优于地高辛。结论:强心冲剂能通过调节某些心肌凋亡基因表达,从而抑制心肌细胞凋亡的发生,这可能是其干预心室重构,防治心力衰竭的机制之一。Objective:This study was designed to making model of heart failure and observed the effect of QiangxinChongji on myocardial apoptosis and ventricular remodeling expression of rabbits with heart failure.Methods: 60 rabbits was randomly divided into 6 groups,each group has 10 rabbits: sham operation group,model group,Digoxin group,low,media,high dose group of QiangxinChongji.Establishing the rabbits model of heart failure by using aorta abdominalis constriction,the course was 9 days.Studying the effects of QiangxinChongji on Bax,Bcl-2,α-actin gene expression.Results: QiangxinChongji could significantly reduce Bax protein expression of rabbits with heart failure,escalated Bcl-2 andα-actin protein expression.Conclusion: QiangxinChongji can inhibit cardiomyocyte apoptosis in rabbits model with heart failure from regulating some gene expression of cardiomyocyte apoptosis,it may be one of the mechanisms of QiangxinChongji to treat heart failure.
