摘  要:目的研究重组人促红细胞生成素(recombinant human erythropoietin,rHuEPO)对急性脊髓损伤大鼠核转录因子-κB(nuclear factor-kappa B,NF-κB)表达的影响。方法参照Nystrom's压迫方法制作大鼠脊髓压迫损伤模型,成年健康Wistar大鼠72只,雌雄不限,按随机数字表法分为正常对照组8只、损伤组32只、重组人促红细胞生成素治疗组32只。术后应用联合行为评分(combined behavioral score,CBS)评价大鼠脊髓神经功能;免疫组化和Western blot检测各组大鼠NF-κB的表达。结果三组CBS评分结果显示,损伤组>重组人促红细胞生成素治疗组>对照组。免疫组化结果显示,损伤组与rHuEPO治疗组NF-κB阳性产物的平均光密度值(MOD)显著高于正常对照组(<0.01),而rHuEPO治疗组与损伤组相比MOD值明显降低(<0.01)。Westernblot结果显示,与正常对照组比较,损伤组与rHuEPO治疗组组NF-κB的灰度值(IDV)与内参照IDV的比值明显升高(<0.01),而rHuEPO治疗组则明显低于损伤组(<0.01)。结论 rHuEPO治疗组很可通过下调NF-κB的表达参与脊髓继发性损伤的修复。Objective To study the effect of recombinant human erythropoietin(rHuEPO)on the expression of nuclear factor-kappa B(NF-κB)in acute spinal cord injury rat.Methods The spinal cord injury was induced with Nystrom's way,the healthy adult Wistar rats(72)were randomly divided into normal control group,spinal cord injury group,recombinant human erythropoietin group on average.The function of rats was determined by combined behavioral score(CBS),and the expression of NF-κB was observed by immunochemistry and western blot methods.Results The CBS score sequence was: spinal cord injury group rHuEPO normal control group.The mean optic density(MOD)of NF-κB positive product increased significantly in acute spinal cord injury group than in normal control by immunochemistry(0.01),but decreased in rHuEPO than in acute spinal cord injury group(0.01).The expression level of NF-κB protein increased significantly in the spinal cord injury group than in normal control(0.01),but decreased in rHuEPO than in spinal cord injury group(0.01)by Western blot.Conclusion The effect of rHuEPO on acute spinal cord injury might be related to downregulation of NF-κB expression
