摘 要:目的了解外侧隔核ghrelin对大鼠胃运动的调节作用及机制。方法大鼠56只,外侧隔核分别微量注射生理盐水(NS)、20nmol/L ghrelin、200nmol/L ghrelin、2 000nmol/L ghrelin、25nmol/L D-Lys3-GHRP-6、NS+25nmol/L D-Lys3-GHRP-6及200nmol/L ghrelin+25nmol/L D-Lys3-GHRP-6混合液(各8只),注射量均为1μL。另取大鼠32只,其中8只单纯切断迷走神经,8只迷走神经切断后外侧隔核微量注射NS 1μL,8只假手术后外侧隔核微量注射200nmol/L ghrelin 1μL,8只迷走神经切断后外侧隔核微量注射200nmol/L ghrelin1μL。采用应力传感器及银丝电极胃内置入方法,记录各组清醒自由活动大鼠胃收缩幅度和频率的变化。结果外侧隔核注射各浓度ghrelin 5min后,胃收缩幅度显著增高(q=3.69~15.93,P<0.05、0.01),胃收缩频率显著加快(q=4.69~15.02,P<0.05、0.01),呈显著剂量依赖关系。外侧隔核注入ghrelin+D-Lys3-GHRP-6混合液5min后,与ghrelin+NS组比较,ghrelin促进胃收缩幅度和频率增加作用消失(t=13.13、3.29,P<0.01),但单用ghrelin受体拮抗剂D-Lys3-GHRP-6对胃收缩幅度和收缩频率均无显著影响(t=0.12、0.28,P>0.05)。膈下迷走神经切断后,大鼠胃收缩幅度和收缩频率显著降低(t=4.39、3.16,P<0.01);与假手术对照组相比,膈下迷走神经切断大鼠外侧隔核注射ghrelin,胃收缩幅度和收缩频率显著降低(q=21.42、6.58,P<0.01),但与迷走神经切断+NS组相比,胃收缩幅度和频率无显著改变(q=0.13、0.16,P>0.05)。结论外侧隔核外源性ghrelin可促进大鼠胃运动,该效应可能通过隔核-迷走通路来完成的。Objective To investigate the effect of ghrelin of lateral septal nucleus(LSN) on gastric motility in rats and its mechanism.Methods Fifty-six rats were evenly divided into eight groups,and a microinjection of 1 μL of the following solutions-i.e.NS,20 nmol/L ghrelin,200 nmol/L ghrelin,2 000 nmol/L ghrelin,25 nmol/L D-Lys3-GHRP-6,NS+25 nmol/L D-Lys3-GHRP-6 and 200 nmol/L ghrelin+25 nmol/L D-Lys3-GHRP-6 was injected into LSN,respectively.Another 32 rats were enrolled,of which,eight underwent merely vagotomy,eight was micro-injected with 1 μL of NS on LSN after vagotomy,eight received 20 nmol/L ghrelin injection on LSN after a sham surgery,and eight were injected with 200 nmol/L ghrelin on LNS after vagotomy.The changes of amplitude of constriction and frequency of gastric motility were measured in conscious rats by a transducer and electrode embedmented into the stomach.Results The rats at 5 min after injection of ghrelin of 20,200 or 2 000 nmol/L showed increased amplitude of constriction(q=3.69-15.93;P〈0.05,0.01) and frequency of gastric motility accelerated(q=4.69-15.02;P〈0.05,0.01),showing an obvious dose-dependent.Compared with the group of ghrelin+NS,the effect of ghrelin on promoting gastric motility,including the amplitudes and frequency(t=3.29,13.13;P〈0.01),significantly disappeared after ghrelin+D-Lys3-GHRP-6 mixture was injected into the LSN.However,there was no significant changes of gastric motility while administration of ghrelin receptor antagonist D-Lys3-GHRP-6 alone(t=0.12,P〉0.05).After vagotomy,the gastric motility amplitude and frequency were significantly decreased.Compared with the sham operation,the amplitude and frequency of gastric motility were significantly reduced after administration of 1 μL of 200 nmol/L ghrelin into LSN of post-vagotomy rats(q=21.42,4.39;P〈0.01),However,there was no significant changes of amplitude and frequency(q=0.13,0.16;P〉0.05) of gastric motility as compared with the NS group.Conclusion Exogenous ghrelin of LSN may
