鲤鱼赤小豆汤对阿霉素肾病大鼠CD2AP及Nephrin表达影响

摘　　要：目的观察阿霉素肾病大鼠肾小球足细胞Nephrin和CD2相关蛋白(CD2AP)的表达及鲤鱼赤小豆汤对其表达的影响。方法 SPF级Wistar雄性大鼠50只,随机分为正常组(N组)10只及造模组40只。造模组一次性尾静脉注射阿霉素6.5mg/kg以制备肾病模型,造模成功后随机分为模型组(M组)、福辛普利组、鲤鱼赤小豆汤低剂量组、鲤鱼赤小豆汤高剂量组。实验干预7周。检测各组大鼠12h尿蛋白、血清总蛋白、清蛋白,免疫组织化学方法检测各组大鼠肾小球内Nephrin及CD2AP的阳性表达水平。结果与N组相比,其他各组12h尿蛋白含量均升高(t=2.38～4.77,P<0.05),尤以M组升高显著。与N组比较,其他各组肾小球中Nephrin与CD2AP的表达减弱(t=3.78～11.06,P<0.05);与M组相比,各干预组肾小球中Nephrin与CD2AP的表达都有所增强(t=2.11～3.48,P<0.05)。结论鲤鱼赤小豆汤可通过上调其肾小球足细胞Nephrin和CD2AP的表达,起到保护肾小球滤过屏障的作用。Objective To observe the expressions of nephrin in podocytes and CD2-associated protein（CD2AP） in adriamycin-induced nephrosis（AIN） in rats,and study the effect of Liyu Chixiaodou Tang（a traditional Chinese decoctions） on them.Methods Fifty male Wistar rats in SPF grade were randomly divided into two groups,10 in normal group（N group） and 40 for pre-model group.A nephrotic model was created in 40 rats in the pre-model group by a single injection of adriamycin（6.5 mg/kg） in tail vein.Upon completion of the models,the rats were divided into model group（M group）,fosinopril group,low-dose Liyu Chixiaodou Tang group,and high-dose Liyu Chixiaodou Tang.The trial lasted for seven weeks.The 12-hour urine protein,total serum protein and total albumin were detected.The expressions of nephrin and CD2AP were measured by immunohistochemistry.Results Compared with the N group,the 12-hour urine protein levels of the other groups were elevated（t=2.38-4.77,P〈0.05）,being especially higher of the M group;the expressions of nephrin and CD2AP in glomerular were decreased（t=3.78-11.06,P〈0.05）.Compared with the M group,the expressions of nephrin and CD2AP were enhanced in each intervention group（t=2.11-3.48,P〈0.05）.Conclusion Liyu Chixiaodou Tang can protect the glomerular filtration barrier by up-regulating the expressions of nephrin and CD2AP.
