摘 要:目的观察磁性纳米粒子对巨噬细胞的吞噬功能有无抑制作用。方法利用透射电子显微镜观察RAW264.7细胞对磁性纳米粒子(二巯基丁二酸修饰的γ-三氧化二铁纳米粒子,DMSA-γ-Fe_2O_3)的摄入情况,流式细胞术检测磁性纳米粒子预处理的RAW264.7细胞吞噬大肠杆菌(E.coli)的吞噬能力。结果 Fe_2O_3磁性纳米粒子能够被RAW264.7细胞摄入,并且随孵育时间延长进入细胞内的磁性纳米粒子增多;与正常RAW264.7细胞相比,磁性纳米粒子预处理的RAW264.7细胞参与吞噬活动的细胞百分比差异无统计学意义(P>0.05)(均约为80%细胞有吞噬活动),但磁性纳米粒子预处理的RAW264.7细胞吞噬荧光标记的E.coli的数量显著减少(P<0.05),而磁性纳米粒子预处理不同时间的RAW264.7细胞其吞噬功能差异无统计学意义(P>0.05)。结论 Fe_2O_3磁性纳米粒子抑制了RAW264.7细胞的吞噬能力。Objective To investigate the influence of superparamagnetic iron oxide nanoparticles ( SPION, or DMSA - γ-Fe2O3 ) on the phagocytosis function of RAW264.7 macrophage - like cells. Methods The internalization of SPION by RAW264.7 cells was observed by transmission electron microscopy (TEM); the phagocytosis function of SPION - loaded RAW264. 7 ceils was measured by flow cytometry. Results More SPION were internalized by RAW264. 7 cells with incubation time extension. Compared with the SPION - unloaded RAW264.7 cells, the percentage of SPION- loaded RAW264. 7 cells which showed phagocytosis activities did not show significant difference. However, the internalization of the fluorescence - labeled E. coli by SPION - loaded RAW264. 7 cells significantly decreased ( P 〈 0.05 ), while no difference of the phagocytosis function among the SPION -loaded RAW264.7 cells with different SPION incubation times. Conclusion SPION depressedthe potential phagocytic capacity of RAW264.7 cells.
