摘 要:目的探讨异丙酚对百草枯(paraquat,PQ)中毒所致肺损伤及核因子-κB(nuclear factor kappaB,NF-κB)在肺组织中表达的影响。方法 128只雄性Wistar大鼠分为3组,对照组(8只),染毒组(60只),异丙酚组(60只)。50mg/kg PQ一次性灌胃建立染毒大鼠模型,对照组1mL/kg生理盐水一次性灌胃。异丙酚组染毒后给予1%异丙酚注射液10mg/kg腹腔注射,2次/d,连用4d;染毒组染毒后予等体积的生理盐水腹腔注射,2次/d,连用4d。观察大鼠1、3、7、14、28d(对照组观察7d)的肺组织病理改变,并用链霉亲和素生物素标记法(labelledstreptavidin biotin method,LSAB)检测NF-κB和肿瘤坏死因子-α在3组大鼠各时间段肺组织中的表达变化。结果染毒组大鼠早期表现为肺水肿、出血及炎细胞浸润,14d后出现肺泡间隔纤维性增厚;异丙酚组上述早期表现明显减轻,但14d后与染毒组比较无明显改善。染毒组大鼠肺组织中NF-κB和肿瘤坏死因子-α于染毒后1、3d时有明显表达,7d后表达明显减弱;异丙酚组NF-κB和肿瘤坏死因子-α在不同时表达与染毒组比较差异无统计学意义(P>0.05)。结论异丙酚能明显减轻PQ中毒大鼠急性期的肺组织损伤,但对NF-κB表达无明显影响。Objective To investigate the influence of propofol on paraquat(PQ) -induced lung injury and the expression of nuclear factor kappaB in lung tissue of rats. Methods A total of 128 male Wistar rats were randomly divided into 3 groups, the control group ( n = 8 ), the intoxication group ( n = 60 ), the propofol group (n = 60 ). Rats in propofol group received PQ (50mg/kg body weight) in a single oral and propofol at a dose of 10mg/kg body weight after PQ, twice daily, intraperitoneally; Intoxication group was treated identically to the propofol Control group received lmL/kg body weight group except for the substitution of 0. 9% saline for propofol ; of saline, orally. Pathological changes of lung were examined under optical microscope on the 1st, 3rd, 7th, 14th and 28th day after intoxication. Meanwhile, the expression of nuclear factor kappaB ( NF - κB ) p65 and tumor necrosis factor - α ( TNF - α) in lung tissue were detected by LSAB immunohistochemistry (IH) staining. Results The histological changes, such as alveolar edema, hemorrhage and inflammatory cell infiltration of the propofol group were milder than those of the intoxication group. The expression of NF - κB p65 and TNF - α in lung tissue of intoxication group and propofol group were similar in that they were both significantly increased on the first day and the 3rd day compared with control group and weakened obviously after the 7th day. Propofolshowed no influence on the expression of both. Conclusion Propofol could relieve paraquat - induced lung injury, but showed no influence on the expression of NF - κB.
