封闭环境对C57BL/6J小鼠听功能及听皮层GABA能神经元凋亡的影响

摘  要:目的探讨封闭环境对老年性聋动物模型C57BL/6J小鼠听皮层GABA能神经元凋亡的影响。方法构建封闭环境,将2、10月龄C57BL/6J小鼠(实验组,各20只)放入其中,并将另2、10月龄C57BL/6J小鼠各20只设立为开放环境对照组,饲养2月后比较不同环境各组动物ABR反应阈变化,免疫荧光法及tunel法检测听皮层GABA能神经元凋亡水平。结果实验组各频率ABR阈值明显高于对照组,尤其以高频明显(P<0.05);tunel检测证实实验组动物听皮层GABA能神经元凋亡显著增多。结论封闭环境能促进老年性聋动物模型C57BL/6J小鼠听皮层GABA能神经元的凋亡,可能是老年性聋的重要环境因素。Objective To evaluats the effect of enclosed environment of on apoptosis of auditory cortes GABA neurons in C57BL/6J mice. Methods Twenty of 2 morths old and 10 months old C57BL/6J mice were ot into an en- closed environment. The same amount and same and age of C57BL/6J mice were out into an open environment as control groups. Two months later, the ABR threshotds were evaluated,and apoptosis of auditory cortex GABA neu- rons were detected by Tunel assny and immunostaining. Results In the mice in the endosed environ meat,the ABR thresholds elevated and the apoptosis were significantly increased. Conclusion Enclosed environment may promote apoptosis of GABA neurons in mile,may be an impotant {actor for presbycusis.
