摘  要:目的探讨α-平滑肌肌动蛋白(α-SMA)阳性细胞(周细胞)在早期心肌缺血中的作用。方法将30只雌性Wistar大鼠皮下多点注射异丙肾上腺素(Isp),对照组注射生理盐水,于0(对照)、4、6、12、24、48h分别麻醉5只大鼠,眼球取血,分离血清,取大鼠心脏,用4%的多聚甲醛溶液固定。生化自动分析仪测定各组大鼠血清天门冬氨酸胺基转移酶(AST)、乳酸脱氢酶(LDH)、肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)水平,采用免疫组织化学染色技术及形态定量分析方法检测心肌组织α-SMA、波形蛋白(vimentin)的表达情况,观察周细胞在心肌缺血早期的变化。结果皮下注射Isp后,12、24、48h组的大鼠血清AsT[(160.25±3.86)、(172.60±8.82)、(192.20±25.35)U/L]、LDH[(1466.254±643.38)、(1645.20±326.83)、(1701.604±640.06)U/L]水平均高于对照组[(129.184±19.65)、(849.454±248.54)U/L,P均〈0.05];4、6、12、24、48h组的大鼠血清CK水平[(1097.60±301.57)、(1247.204±243.84)、(1263.754±271.22)、(1448.004±647.00)、(1268.40±479.81)U/L]、CK—MB水平[(217.124±35.89)、(229.08±97.11)、(251.704±46.82)、(318.804±77.76)、(249.044±98.54)U/L]均高于对照组[(713.454±146.30)、(147.054±25.75)U/L,P均〈0.05];4h组大鼠心肌周细胞的数量[(61.00±17.25)个]组高于24、48h组[(28.20±5.81)、(t8.20±2.17)个],48h组低于6h组[(50.004±3.61)个.P均〈0.05]。6、12、24、48h组大鼠心肌vimentin表达水平(4.174±2.49、5.244±2.84、8.374±2.18、7.734±2.49)均高于对照组和4h组(1.884±1.85、2.21±1.54,P均〈0.05),24、48h组均高于6、12h组(P均〈0.05)。结论缺血早期周细胞的数量多于其他间质细胞,周细胞�Objective To study the role of alpha smooth muscle actin(α-SMA) positive cells(pericytes) in early myocardial ischemia. Methods Thirty healthy female Wistar rats were randomly divided by body weight into normal control group and subcutaneous multi-point injection of isoprenaline(Isp) group. Five rats were anesthetized after 0, 2, 4, 6, 12, 24 and 48 h in each group. Blood was taken in eyeballs and, serum was separated, heart was taken and fixed in 4% paraformaldehyde solution. The myocardial enzymes [serum apartate aminotransferase (AST), lactate dehydrogenase (LDH), creatine kinase (CK), creatine kinase isoenzymes (CK-MB)] were determined by biochemical automatic analyzer, the expression of ct-SMA and vimentin in myocardial tissue was detected by immunohistochemical staining and changes of pericytes in early myocardial ischemia was observed by morphometric analysis. Results The level of myocardial enzymes AST[(160.25 ± 3.86), (172.60 ± 8.82), (192.20 ± 25.35) U/L], and LDH[(1466.25 ± 643.38), (1645.20 ± 326.83), (1701.60 ± 640.06)U/L], 12, 24 and 48 h after subcutaneous injection of Isp, were higher than that[ (129.18 ± 19.65), (849.45 ± 248.54)U/L, all P 〈 0.05] of the control group. The level of CK[(1097.60 ± 30157), (1247.20 ± 243.84), (1263.75 ± 271.22), (1448.00 ± 647.00), (1268.40 ± 479.81)U/L], and CK-MB[(217.12 ± 35.89), (229.08 ± 97.11), (251.70 ± 46.82), (318.80 ±77.76), (249.04 ± 98.54)U/L], 4, 6, 12, 24 and 48 h after subcutaneous injection of Isp, were higher than that [(713.45 ± 146.30), (147.05 ± 25.75)U/L, all P 〈 0.05] of the control group. The number of ct-SMA positive cells(61.00 ± 17.25), 4 h after Isp injection, was significantly increased compared with that(28.20 ± 5.81, 18.20 ± 2.17) of 24 and 48 h after Isp injection. The number of ct-SMA positive cells in 48 h group was less than that(50.00 ± 3.61, P 〈 0.05) of 6 h group. The number of vim
