摘 要:目的:观察黄芪皂甙和丹参酮ⅡA对大鼠骨骼肌急性钝挫伤后胚胎型肌球蛋白重链(embryonic Myosin Heavy Chain,MHC-emb)和波形蛋白(vimentin)表达的影响。方法:80只雄性Wistar大鼠(体重180~200g)随机分为A组(黄芪皂甙组)、B组(丹参酮ⅡA组)、C组(黄芪皂甙+丹参酮ⅡA组)、D组(黄芪丹参注射液组)、E组(生理盐水对照组),每组16只。采用打击装置造成右侧腓肠肌中段钝挫伤模型,分别于损伤局部注射相应药物,并于损伤后4、7、14和28天进行腓肠肌损伤部位取材。Western Blotting方法检测各组MHC-emb和波形蛋白表达量的变化。结果:①与生理盐水组相比,各干预组MHC-emb表达均明显升高,差异有统计学意义(P<0.05),这种情况一直持续到伤后28天;其中又以C组和D组升高最为明显;②与生理盐水组相比,各干预组波形蛋白表达均明显降低,差异有统计学意义(P<0.05),这种情况一直持续到伤后28天;其中又以C组和D组降低最为明显。结论:在急性骨骼肌钝挫伤修复过程中,黄芪皂甙、丹参酮ⅡA均能促进MHC-embmRNA表达,抑制波形蛋白mRNA表达,联合使用时效果最佳,与黄芪丹参复方成分基本一致。Objective To observe the effects of astragaloside and tanshinone ⅡA on the expression of embryonic Myosin Heavy Chain(MHC-emb) and vimentin following acute skeletal muscles contusion in rats.Methods 80 male Wistar rats(weight 180-200 g) were divided into group A(Astragaloside),group B(Tanshinone ⅡA),group C(Astragaloside and Tanshinone ⅡA),group D(Huangqi-Danshen) and group E(physiological saline control) randomly,16 rats in each group.Right gastrocnemii were contused in all groups by specific device,and then the damaged locations were injected with the corresponding drug.The samples were collected from injured muscles 4,7,14 and 28 days after injury.Western-Blotting analysis was used to measure the expression of MHC-emb and vimentin in each group at different time point.Results ① Compared to the group of physiological saline control,the expression of MHC-emb in each intervention group increased significantly(P〈0.05) after injury,which continues until 28 days after injury;The expression of MHC-emb increased most obviously in group C and group D.② Compared to the group of physiological saline control,the expression of vimentin in each intervention group decreased significantly(P〈0.05) after injury,which continues until 28 days after injury;The expression of vimentin decreased most obviously in group C and group D.Conclusion During the repairing process of acute contusion of skeletal muscle,Astragaloside and Tanshinone IIA promoted the expression of MHC-emb and inhibited the expression of vimentin.The effect of mixed injection of Astragaloside and Tanshinone ⅡA was similar to the Huangqi-Danshen injection.
