大黄水提物对雄性小鼠睾丸影响的实验研究

摘　　要：目的:观察大黄水提物对雄性小鼠睾丸的影响。方法:40只8周龄雄性昆明种小鼠随机分为空白对照组、大黄水提物大、中、小剂量组,每组10只小鼠,大黄三个剂量组小鼠分别给予2g/kg、1g/kg、0.5g/kg的大黄水提物连续灌胃30天。分别计算各组小鼠的睾丸指数,HE染色观察睾丸的形态结构。结果:与空白对照组比较,大黄中、大剂量组小鼠的睾丸指数明显降低(P<0.01,P<0.05);且大剂量组小鼠的睾丸指数明显低于小剂量组(P<0.05)。HE染色结果显示,大黄三个剂量组小鼠睾丸的形态结构出现明显的病理变化,以大剂量组最为严重。结论:长期应用大黄可损害睾丸的形态结构,以大剂量的损害最为严重。Objective：To observe the toxic effects of rheum aqueous extract on male mice testes.Methods：Forty 8-week male Kunming mice were randomly divided into 4 groups （n=10）： blank conlrol group and rheum high, middle, low dose group. The mice in rheum groups were respectively lavaged with 2g/kg, lg/kg and 0.5g/kg rheum aqueous extract for 30 days. The testicular index of mice in each group was calculated; HE stain was used to observe morphology of testes.Results：Compared with blank control group, the testicular index of mice in rheum middle, high dose group obviously decreased （P〈 0.01, P〈 0.05）; moreover, the testicular index of mice in rheum high dose group wereobviously lower than low dose group （t9〈 0.05）. The morphology of mice testes in 3 rheum groups appeared obvious pathological changes, and the high dose group was the worst. Conelusions：Long-term using of rheum can damage the morphology of mice testis, and the injury of high dose rheum is the most serious.
