摘 要:目的:探讨前列地尔对局灶性脑缺血模型大鼠大脑皮层中诱导型一氧化氮合酶(iNOS)和内皮型一氧化氮合酶(eNOS)表达的影响。方法:取大鼠随机分为假手术组、模型组、阳性对照(尼莫地平1mg.kg-1)组和前列地尔低、中、高剂量(1.25、2.50、5.00μg.kg-1)组,每组10只,腹腔注射相应药物6d,每日1次,末次给药后建立局灶性脑缺血模型,大鼠清醒后进行神经功能缺损评分,采用免疫组化法和免疫印迹法检测各组大鼠建模2h后iNOS、eNOS的表达。结果:与假手术组比较,其余各组神经功能缺损评分均明显增加,iNOS阳性细胞数和表达量均明显增加,除模型组外其余各组eNOS阳性表达量均增加(P均<0.05);与模型组比较,阳性对照组和前列地尔中、高剂量组神经功能缺损评分均明显降低,阳性对照组和前列地尔3个剂量组iNOS阳性细胞数和表达量均明显降低、eNOS阳性细胞数和表达量均明显增加(P均<0.05)。结论:前列地尔预处理对局灶性脑缺血模型大鼠具有保护作用,其机制可能与下调大脑皮层iNOS、上调eNOS的表达有关。OBJECTIVE: To study the effects of alprostadil on the expressions of inducible nitric oxide synthase (iNOS) and endothlial nitric oxide synthase (eNOS) in cerebral cortex of rats with focal cerebral ischemia. METHODS: Rats were randomly di- vided into the sham operation group, model group, positive control group (nimodipine 1 mg.kg ') and alprostadil high-dose, medi- um-dose and low-dose groups (1.25, 2.50, 5.00 ~tg. kg-~) with 10 rats in each group. Those groups were given relevant medicine once a day for 6 days. Cerebral ischemia models were made after the last administration. After being awake, the neurological deficit score was evaluated. Immunohistochemistry method and Western blot technique were used to observe the expressions of iNOS and eNOS 2 h after modeling. RESULTS: Compared with sham operation group, the neurological deficit scores, the amount and expres- sion of iNOS were increased significantly in other groups; the expression of eNOS was significantly increased in those groups ex- cept for model group (P〈0.05). Compared with model group, neurological deficit scores of positive control group, alprostadil me- dium-dose and high-dose groups were decreased significantly, and the amount and expression of iNOS in positive control group and alprostadil groups were decreased significantly while the amount and expression of eNOS were increased significantly (P〈0.05). CONCLUSION: Alprostadil can protect the brain cells from damage after focal cerebral ischemia, which may be associated with down-adjusting the expression of iNOS and up-adjusting the expression of eNOS.
