摘 要:目的对五年生人参根生长发育过程中苹果酸脱氢酶(MDH),乳酸脱氢酶(LDH),乙醇脱氢酶(ADH),葡萄糖-6-磷酸脱氢酶(G6PDH),α-磷酸甘油脱氢酶(α-GPD)5种脱氢酶的活力进行比较。方法分别于展叶期、开花期、结果期、果熟期、枯萎期采取人参根样品;采用中性磷酸缓冲液提取粗酶液;应用分光光度法测定MDH、LDH、ADH、G6PDH、α-GPD的活力。结果在人参根生长发育过程中,MDH、ADH、G6PDH活力均逐渐升高;LDH活力在结果期和果熟期分别出现活力峰值;α-GPD在展叶期和果熟期末期活力最高。结论 MDH、LDH、ADH、G6PDH、α-GPD的活力比较可作为判断人参生长发育状况的评价指标。AIM To compare the activities of malate dehydrogenase ( MDH), lactate dehydrogenase ( LDH), alcohol dehydrogenase (ADH), glucose-6-phosphate dehydrogenase (G6PDH) and α-glycerophosphate dehydro- genase (α-GPD) in Ginseng Radix et Rhizoma during five growth and development period. METHODS Ginseng Radix et Rhizoma samples were collected from the leaf period, the flowering period, the fruit setting period, the fruit ripening period and the withering period. Neutral phosphate buffer solution was used to extract the enzyme so- lution of Ginseng Radix et Rhizoma. The activities of MDH, LDH, ADH, G6PDH and α-GPD were tested by spec- trophotometry. RESULTS In the growth and development process of Ginseng Radix et Rhizoma, the activity of MDH, ADH, G6PDH gradually increased, there were two peaks of the activity of LDH in fruit setting period and the fruit ripening period, α-GPD had the highest activity in the leaf period and the end of the fruit ripening period. CONCLUSION The comparison among the activities of MDH, LDH, ADH, G6PDH, α-GPD could be used as indictors for the growth and development period.
