摘 要:目的采用加速溶剂萃取方法提取油菜籽饼粕中原花青素,利用响应面分析法对其提取工艺参数进行优化。方法以脱脂油菜籽粉末为原料,乙醇作为提取剂,研究了乙醇浓度、萃取温度和静态循环次数3个因素对原花青素提取量的影响、确定加速溶剂萃取法最佳提取条件;采用紫外分光光度法测定其含量,并与超声提取法进行了对比。结果油菜籽饼粕中原花青素的最佳提取条件是乙醇浓度60%、萃取温度60℃、静态萃取次数3次。在此条件下,加速溶剂萃取法所提取的原花青素的含量是7.370 mg/g,而超声法提取原花青素含量为6.812 mg/g。结论加速溶剂萃取技术具有省时、方便、提取效率高等优点,适合于油菜籽饼粕中原花青素的提取。Objective Accelerated solvent extraction was employed to extract proanthocyanidins from rapeseed meal,using response surface methodology to optimize the parameters of its extraction process. Methods Defatted rapeseed and ethanol were selected for raw material and extractant.Concentration of ethanol,extraction temperature,and static cycle times were studied to determine the effect on proanthocyanidins and optimize conditions.Its contents were determined by UV spectrophotometry compared with the ultrasonic extraction. Results The best extraction conditions of accelerated solvent extraction determined by response surface were as follows: concentration of ethanol 60 %,extraction temperature 60 ℃,cycle number 3 times.Under optimum conditions,the content of procyanidins was 7.370 mg/g,but the content of procyanidins with ultrasonic extraction was 6.81 mg/g. Conclusion Compared with ultrasonic extraction method,ASE is time-saving,convenient,highly efficient and it is suitable for extraction of proanthocyanidins from rapeseed meal.
