摘 要:目的观察两种氧化剂对阿萨希毛孢子菌(T.asahii)临床分离株与环境分离株生长发育的影响。方法临床分离株与环境分离株菌悬液分别接种于加入了不同浓度的过氧化氢和甲萘醌的马铃薯葡萄糖琼脂培养基(PDA培养基)与酵母浸出粉胨葡萄糖液体培养基(YPD液体培养基)中,肉眼、光镜及电镜下观察菌株的形态变化。结果①肉眼形态,随氧化剂浓度的升高,菌落直径逐渐缩小,当达到一定浓度时,菌落不再生长;相同条件下,环境分离株较临床分离株菌落直径小;②光镜形态,菌落发育延迟,数量减少,见许多芽管,随氧化剂浓度的升高,出现许多不易着色、发育不全的孢子,体积较小,偶见胞质萎缩的稍大孢子;③电镜形态,两种氧化剂对两株菌均有损伤,可见菌体表面皱缩,胞壁部分至完全破损,胞质溢出,菌株体积缩小。结论两种氧化剂对两株菌生长发育均有影响,呈浓度依赖,但甲萘醌对菌株影响更强;环境分离株受氧化剂影响更大。Objective To observe hydrogen peroxide and menadione on Trichosporon asahii (Zasahii)isolates obtained from clinical and environmental growth and development in vitro. Methods Growth of cultures was monitored in potato-dextrose agar (PDA) medium and yeast extract-peptone-dextrose (YPD) liquid medium containing increasing concentrations of the above oxidants, observing cell and colony morphologies from the whole, light microscopy(LM) and scanning electron microscopy(SEM). Results On the whole, compared with the control,the colony diameter gets smaller and smaller with the oxidant concentration increased, when a certain concentration, the strain will not grow under the same conditions. Environmental isolates were smaller than clinical isolates in colony diameter. Form of light microscopy, strains affected by oxidants development delay and the number reduce, there are a number of germ tubes. With the oxidant concentration increased, spores colordifficult, hypoplasia, smaller, and occasionally cytoplasm shrinking in larger spores. In electron microscopy: showing that cell surface shrinkage, part of the cell wall to completely damaged, cytoplasmic overflow, strains smaller in size. Conclusion Between the two oxidants tested, menadione had a comparatively higher damaging effect. And environmental isolates are more sensitive to the oxidant.
