摘 要:以毛尖紫萼藓(Grimmia pilifera)为试材,采用RACE技术克隆获得1个抗坏血酸过氧化物酶基因(APX)的cDNA序列,命名为GpAPX(GenBank登录号:GU989311)。该基因全长1 115 bp,编码区为771 bp,编码256个氨基酸,5'非翻译区为116 bp,3'非翻译区为228 bp。序列分析表明:该基因属于过氧化物酶基因家族,与其他植物抗坏血酸过氧化物酶的同源性为70.2%~89.7%。实时定量PCR检测结果显示,GpAPX在干旱及复水条件下均能表达,表明GpAPX蛋白可能与抗旱性相关,在毛尖紫萼藓响应干旱胁迫过程中发挥作用。In this study,a full-length cDNA sequence of a ascorbate peroxidase(APX)gene named GpAPX was cloned from Grimmia pilifera(GenBank accession No.GU989311)using rapid amplification of cDNA ends(RACE).The full-length cDNA of GpAPX was 1 115 bp with a 771 bp open reading frame(ORF)which encoded 256 amino acids and containing a 116 bp 5′UTR and a 228 bp 3′UTR.Sequence analysis showed that GpAPX belonged to the peroxidase protein families.GpAPX protein sharing 70.2%-89.7% homology with APX proteins from other plants.QRT-PCR analysis suggested that the expression of GpAPX gene was induced in both dehydration and rehydration.The results demonstrated that the GpAPX protein may be involved in drought resisitance,and play an important role in Grimmia pilifera through responses to drought stress.
