高效液相色谱法测定盐酸左旋多巴异戊酯的含量

摘  要:目的建立测定盐酸左旋多巴异戊酯含量的高效液相色谱法。方法色谱柱采用UltimateTMC18柱(250 mm×4.6 mm,5μm),流动相为0.05 mol/L磷酸二氢钾(磷酸调pH=3.5)-乙腈(70∶30),流速为1.0 mL/min,检测波长为280 nm,进样量为20μL,柱温为30℃。结果盐酸左旋多巴异戊酯质量浓度在16.1~322.0μg/mL范围内与峰面积线性关系良好(r=0.999 9),平均回收率为98.41%,RSD为1.85%(n=6)。结论该方法准确、简便,精密度高、专属性强,可用于盐酸左旋多巴异戊酯的含量测定。Objective To establish a high performance liquid chromatography(HPLC) method for the content determination of levodopa isopentyl ester hydroehloride. Methods The Ultimate C18 column (250mm×4.6mm,5μm) was used and the mobile phase was acetonitrile was 0.05 mol/L potassium dihydrogen phosphate (adjusting pH 3.5 with phosphoric acid, 70 : 30), the flow rate was 1.0 mL/miu. The detection wavelength was 280 nm. The column temperature was 30 ℃ and the injection volume was 20μL. Results All the related substances gained a completely chromatographic separation with the principal peak. A good linear relationship was observed in the concentration range of 16.1-322.0 μg/mL, with the correlation coefficient of 0.999 9(n=6). The average recovery rate was 98.41% and RSD was 1. 85% (n =6). Conclusion The method is proved to be accurate and sensitive with high precision and strong specificity, which can be applicable for the content assay of levodopa isopentyl ester hydrochloride.
