摘 要:以含有1 mmol/L Al3+的S-LB培养基作为筛选培养基,从酸性土壤中分离到13株耐铝的细菌菌株,选取其中6株进行形态学分析,结果观察到这些菌株的菌体均呈杆状,其中1株为革兰阳性反应,其余5株为革兰阴性反应。以细菌通用引物扩增这些菌株的16S rDNA并测序,将得到的序列与GenBank中的序列进行BLAST比对,利用MEGA 4.0软件,按照Neighbor-joining法构建系统进化树,这6个菌株分别与Enterobacter endosymbiont、Serratia marces-cens、Pantoea agglomerans、Enterobacter aerogenes、Bacillus subtilis和Enterobacter asburiae的亲缘关系最近。将这些菌株接种到加有2 mmol/L Al3+、pH 4.5的S-LB固体培养基上培养时,它们都能生长,说明这些菌株具有较好的耐铝能力,这些菌株为进一步研究细菌的耐铝机制提供了极好的材料。Thirteen aluminum (Al) -tolerant bacteria strains were screened and isolated from acid soils using the S-LB medium containing 1 mmol/L Al3 +. Of the thirteen strains, six were selected to perform morphological analysis. The results in- dicated that the cells of all these strains were rod. One had Gram-positive reaction while the other five had Gram-negative reactions. 16S rDNAs from these strains were amplified with general primers of bacteria to conduct sequence analysis. The obtained sequences were compared with the known sequences in GenBank. The phylogenetic tree was constructed with MEGA 4.0 soft- ware according to Neighbor-joining method. The result showed that the isolated strains were closest relative with Enterobacter endosymbiont, Serratia marcescens, Pantoea agglomerans, Enterobacter aerogenes, Bacillus subtilis and Enterobacter asburiae, respectively. All these strains could grow on LB solid medium ( pH 4.5) containing 2 mmol/L Al3 + , suggesting that they had ability to tolerate higher concentrations of Al3 +. These strains will be very good materials for the future researches on Al-toler- ant mechanism in bacteria.
