摘 要:目的探讨肾心宁方干预AT1受体敲除(AT1-KO)小鼠慢性肾功能不全心室重塑的作用。方法运用AT1-KO小鼠进行5/6肾大部切除法造模,予肾心宁方干预治疗12周,并与单纯肾切除小鼠进行对比,观察血BUN、血Scr、血脑利钠肽(BNP)、心超[左室收缩末期直径(LVDD)、左室舒张末期直径(LVDS)、短轴缩短率(FS)、射血分数(EF)]、心肌形态和超微观结构变化。结果肾心宁方可以在一定程度上降低CRF小鼠的BUN、Scr,降低LVDD、LVDS、BNP,提高FS、EF,使心肌线粒体数量增加、超微结构改善。结论①RASS系统在慢性肾功能不全心室重塑的过程中具有重要作用,拮抗RASS可以有效改善心室重塑;②肾心宁方可以有效改善CRF的肾功能,并与RASS拮抗具有协同作用;③肾心宁方可以有效改善CRF心室重塑,其作用机制与改善RASS、降低尿毒症毒素和改善心肌超微结构相关。Objective To explore the intervene effect of "Shenxinning Prescription" on ventricular remodeling in AT1-KO rats with chronic renal insufficiency. Methods Establish the models by cutting off 5/6 kidney in AT1 -KO rats, treated with "Shenxinning Prescription" for 12 weeks, and compared with simple nephreetomy rats. The BUN, Set, BNP, LVDD, LVDS, FS, EF, changes of myocardium and ultra micro structure were observed. Results "Shenxinning Prescription" can decrease the BUN, SCR, LVDD, LVDS and BNP, and increase the FS and EF to improve the quatity and structure of mitochondria. Conclusion RASS system plays an important role in ventricular remodeling of chronic renal insufficiency, RASS antagonism can improve the ventricular remodeling;" Shenxinning Prescription" is effective to improve the kidney function of CRF, and has synergic action with RASS antagonism; Shenxinning Prescription" is effective to improve the ventricular remodeling of CRF, and the mechanism is related to RASS improvement, uremic toxin reduction and improvement of myocardial ultra micro structure.
