摘 要:小麦WAG1基因是MADS-box家族C功能基因的一种,对小麦心皮和雌蕊的发育起到重要的调控作用.为了解WAG1基因对小麦三雌蕊突变体(TP)形态构建产生的影响,本文利用RT-PCR技术从三雌蕊突变体近等基因系CSTP幼穗中克隆了WAG1基因完整的编码区序列,对其cDNA及推导的氨基酸序列进行分析,结果显示该基因编码区长765bp,编码254个氨基酸,序列与雌蕊化异质小麦(AB084577.1)完全一致;通过定量PCR分析WAG1在CS和CSTP中的差异表达,结果显示该基因在CSTP幼穗不同发育阶段的表达量为:幼穗长度2-5mm﹥5-7mm﹥7-10mm,并且高于在CS幼穗对应阶段的表达量,约是其4.5倍.这表明WAG1基因可能在小麦三雌蕊形成的过程中,尤其是在其雌蕊原基分化发育时期起到了重要作用.本研究的结果对于进一步明确WAG1基因的结构和功能,尤其是在小麦三雌蕊形态构建中的作用提供了基础资料.AGAMOUS of wheat(WAG1) is a class C gene of the MADS-box genes family which plays important roles in flower development and organ differentiation in plants.To study the molecular mechanism of the three pistils mutation in wheat,we cloned and characterized the cDNA sequence of WAG1.The expression patterns of WAG1 were analyzed through real-time quantitative RT-PCR in different developmental stages of young spikes.The open reading frame(ORF) of WAG1 was 765 bp,encoding 254 amino acids.The acquired sequence and the amino acids were the same as what cloned from Alloplasmic Wheat.Expression analysis indicates that young spikes contain higher levels of WAG1 transcripts in the earlier developmental stages.The transcript levels in the young spikes of Chinese Spring three pistils line have increased 4.5-times compared with those in the young spikes of the Chinese Spring line.The data provide a basis for further research on the function of WAG1,and a rationale for the molecular mechanism of the three pistil mutation in wheat.
