国产单硝酸异山梨醇酯注射剂对家兔多种离体血管平滑肌的影响

摘　　要：目的 :研究国产单硝酸异山梨醇酯注射剂对多种离体血管平滑肌的影响。方法 :观察该药物对去甲肾上腺素及氯化钾分别引起家兔多种离体血管平滑肌收缩活动的影响。结果 :单硝酸异山梨醇酯对2×10-6mol·L 1NE引起兔离体降主动脉、肺动脉、肾动脉、股动脉螺旋条及肾静脉、股静脉、门静脉血管条收缩呈浓度依赖性抑制作用 ,其半数抑制浓度依次为(5 5±0 6、2 6±0 1、3 0±0 1、8 7±0 5、0 4±0 1、0 5±0 1、10 1±1 4)μg·mg-1。而该药对40mmol·L-1氯化钾所引起的上述血管条收缩则影响不明显。结论 :国产单硝酸异山梨醇酯注射剂具有明显的扩张血管平滑肌作用 。Objective: To study the effects of native isosorbide 5-mononitrate (5-ISMN) injection on various isolated vascular strips. Methods: To observe the effect of 5-ISMN on both NE- and KCl-induced contraction of various isolated vascular strips in the rabbits. Results: The result showed that 5-ISMN could concentration-dependently restrain the constriction of NE-induced thoracic artery, pulmonary artery, renal artery, femoral artery, renal vein, femoral vein and portal vein strips, with IC50 5.5±0.6, 2.6±0.1, 3.0±0.1, 8.7±0.5, 0.4±0.1, 0.5±0.1, 10.1±1.4 ug.ml-1, respectively. But not by KCl-induced. Conclusion: 5-ISMN injection has a significant dilating vascular effect which more potently in vein than in artery.
