益阳活血方对损伤兔窦房结组织HCN4蛋白表达的影响

摘　　要：目的通过研究中药复方益阳活血方对损伤的兔窦房结组织超级化激活环核苷酸门控阳离子通道4(Hyperpolarization-activated cyclic nucletide-gated cation channel 4,HCN4)表达的影响,探讨其治疗病态窦房结综合征的可能机制。方法取60只大耳白兔(体质量2.3～2.8 kg),随机分为正常组,模型组,阿托品组,益阳活血方高、中、低剂量组,每组10只,除正常组不造模外,余各组采用甲醛加压注射渗透法建立兔窦房结组织损伤模型,模型稳定后开始给药,疗程(7 d)结束后取材,免疫组织化学方法观察窦房结HCN4表达的变化。结果免疫组化染色示窦房结HCN4表达主要位于结中央区,向周边逐渐减少。与正常组相比,造模后各组家兔窦房结组织HCN4蛋白表达均有不同程度降低(P<0.05或P<0.01)。与模型组比较,各用药组IOD值均有不同程度升高,差异有统计学意义(P<0.01),且高剂量组明显优于中、低剂量组及阿托品组(P<0.01),中、低剂量组与阿托品组比较,差异无统计学意义(P>0.05)。结论益阳活血方可提高损伤兔窦房结组织HCN4蛋白的表达,可能是治疗病态窦房结综合征的机制之一。Objective To study the effects of the Yiyang huoxue recipe（YHR）on the expression of hyperpolarization-activated cyclic nucletide-geted cation channel 4（HCN4）in damaged sinoatrial node（SAN）cell in rabbits,and discuss the mechanism of YHR for curing sick sinus syndrome.Methods Sixty healthy adult big-ear white rabbits were randomly divided into 6 groups：the normal group,the model group,the atropine group,and the high/middle/low-dose YHR group,with ten rabbit in each group.The rabbit models of different groups were established by injecting formaldehyde to SAN except the normal group.After the model was established,the rabbits received intragastric administration for 7 d.The expression of HCN4 in SAN was determined by immunohistochemistry.Results Immunohistochemical staining showed that the expression of HCN4 located in the central area of SAN and gradually decreased from the center to borderline.Compared with the normal group,the expression of HCN4 in SAN reduced significantly in other groups（P0.05 or P0.01）.After treated with medicine,the IOD in different groups changed in different degrees,there were significant differences between the model group and the other groups（P0.01）,and the high-dose YHR group were superior to middle-dose YHR group,low-dose YHR group and atropine group（P0.01）,there was no significant difference among the middle-dose YHR group,the low-dose YHR group and the atropine group（P0.05）.Conclusion YHR could improve the expression of HCN4 in SAN,and it could be one of the mechanisms of treating sick sinus syndrome.
