高同型半胱氨酸血症家兔血栓前状态分子标志物的变化研究

摘　　要：目的观察高同型半胱氨酸血症(hyperhomocysteinemia,HHcy)家兔的同型半胱氨酸(homocysteine,Hcy)水平与血栓前状态(prethrombotic state,PTS)指标之间的关系。方法将22只新西兰雄性家兔随机分为两组,正常对照组11只用普通饲料喂养,高蛋氨酸组11只用2%蛋氨酸饲料喂养,自由饮水。0、4、8周心脏采血检测Hcy、血浆血管性血友病因子(von Willebrand factor,vWF)、P-选择素(P-selectin)、血栓前体蛋白(thrombus precursor protein,TpP)含量。结果蛋氨酸组家兔血浆vWF、P-selectin、TpP明显升高,与对照组比较差异有统计学意义(P<0.01)。HHcy家兔的vWF、P-Selectin、TpP水平与Hcy水平之间,均呈明显的正相关(P<0.01)。结论 HHcy家兔存在明显的血栓前状态,表现为继发性纤溶亢进,血小板活性增加及血管内皮细胞损伤。Objective To investigate the relationship between the high homocysteine levels and prothrombotic state in hyperhomocysteinemia（HHcy） rabbits.Methods 22 New Zealand male rabbits were randomly divided into two groups,the normal control group（11 cases） fed with normal diet and the high-methionine group（11 cases） fed with 2% methionine diet,drinking water freely.In 0,4,8 weeks,cardiac blood was collected for testing Hcy,plasma Von Willebrand factor（vWF）,P-selectin,thrombus precursor protein,content（TpP）.Results Plasma vWF,P-selectin and TpP in the high-methionine group group were significantly increased compared with the control group with significant difference（P0.01）.In HHcy rabbits,vWF,P-selectin and TpP levels with the Hcy levels showed a significant positive correlation（P0.01）.Conclusion Obvious prothrombotic state exists in HHcy rabbits,expressing as secondary hyperfibrinolysis,increased platelet activity and vascular endothelial cell injury.
