摘  要:目的探讨原代培养乳鼠窦房结细胞与心房肌细胞中缝隙连接蛋白(connexin,Cx)的表达及差异。方法取培养48h的乳鼠窦房结细胞,随机分为Cx45组、Cx43组、Cx40组;取培养48 h的乳鼠心房肌细胞,随机分为Cx45组、Cx43组、Cx40为对照组,通过激光共聚焦显微镜检测荧光分布及强度变化。结果乳鼠窦房结细胞中Cx45表达明显,Cx43表达次之,而Cx40表达较弱;乳鼠心房肌细胞中Cx40表达明显,Cx45及Cx43表达较弱,乳鼠窦房结细胞Cx45免疫信号比心房肌细胞中高7.8倍(P<0.01),而Cx40的免疫信号则较心房肌细胞低4.0倍(P<0.01)。结论原代培养乳鼠窦房结细胞以Cx45表达为主,而心房肌细胞主要是Cx40表达。Objective To discuss the expression of connexin(Cx) in sinoatrial node cells and atrial muscle cells in neonatal rats.Methods The sinoatrial node cells of neonatal rats cultured for 48 hours were collected and randomly divided into Cx45 group,Cx43 group and Cx40 group.The atrial muscle cells of neonatal rats cultured for 48 hours were collected and randomly divided into corresponding control groups.The changes of fluorescence distribution and intensity were detected by using laser confocal microscope.Results In sinoatrial node cells,the expression of Cx45 was significant,that of Cx43,secondary,and that of Cx40,slight.In atrial muscle cells,the expression of Cx40 was significant and expressions of Cx45 and Cx43 were slight.The immune signal of Cx45 in sinoatrial node cells was 7.8 fold higher than that in atrial muscle cells(P0.01),and immune signal of Cx40 in sinoatrial node cells was 4.0 fold lower than that in atrial muscle cells(P0.01).Conclusion The expression of Cx45 is dominant in sinoatrial node cells and expression of Cx40 is dominant in atrial muscle cells.
