摘  要:研究岩藻甾醇对人早幼粒白血病HL-60细胞的生长及细胞凋亡的影响.采用MTT比色法测岩藻甾醇对HL-60细胞增殖的抑制作用;用Hoechst33258染液染色,在荧光显微镜下观察细胞形态学改变;采用流式细胞术检测细胞周期分布和细胞的凋亡率;比色法检测caspase-9,caspase-3活性.岩藻甾醇能抑制HL-60细胞的生长.MTT法测得岩藻甾醇的IC50为14.2μg/mL.在荧光显微镜下观察药物作用24 h的细胞生长密度逐渐变疏,细胞变小,表面起皱,高质量浓度组大部分细胞破碎.流式细胞仪检测表明,用质量浓度为28.4、14.2、7.1μg/mL的岩藻甾醇作用24 h均出现亚二倍体凋亡峰,凋亡率分别为17.15%、16.42%、15.65%.比色法结果表明岩藻甾醇对HL-60细胞作用24 h后caspase-9、caspase-3的相对活性升高,质量浓度为7.1、14.2、28.4μg/mL的岩藻甾醇对HL-60细胞作用24 h后,caspase-9相对活性分别为135.10%、228.39%、467.74%,与阴性对照组比较有显著性差异(P<0.01).caspase-3相对活性分别为131.06%、224.23%、534.64%,与阴性对照组比较有显著性差异(P<0.01).岩藻甾醇可以抑制HL-60细胞的生长,诱导HL-60细胞凋亡.To investigate the growth inhibiting and apoptosis effect of fucosterol on human promyelocytic leukemia HL-60 cells.MTT assay measured fucosterol on HL-60 cells proliferation;with Hoechst33258 dye staining,cells were observed under a fluorescence microscope morphological changes;flow cytometry cell cycle and cell wither death rate;colorimetric assay caspase-9,caspase-3 activity.Fucosterol can inhibit the growth of HL-60 cells.Fucosterol 1measured by MTT IC50 of sterol 14.2 μg/mL.Observed under a fluorescence microscope 24 h of drug gradually becomes sparse density of cell growth,cells become smaller,surface wrinkling,the majority of high concentration of cell disruption.Flow testing showed that,with a concentration 28.4,14.2,7.1 μg/mL of the fucosterol for 24 h were hypodiploid apoptotic peak appeared,the apoptosis rates were 17.15%,16.42%,15.65%.Assay results showed that concentrations of dose group of 7.1,14.2,28.4 μg/mL of fucosterol on HL-60 cells 24 h after the caspase-9,caspase-3,of the relative activity with the increase of drug concentration increased,caspase-9 relative activity were 135.10% 228.39%,467.74 %,caspase-3 relative activity were 131.06%,224.23%,534.64%,with the negative control group were significantly differences(P0.01).Fucosterols can inhibit the growth of HL-60 cells,induced apoptosis in HL-60 cells.
