双葛解酲丸对酒精性脂肪肝大鼠炎症及脂质代谢的影响

摘　　要：目的探讨双葛解酲丸对酒精性脂肪肝模型的治疗效果。方法将65只SD大鼠分为空白对照组(11只)和酒精性脂肪肝(alcoholic fatty liver,AFL)组(54只),后者给予白酒灌胃建立AFL模型。8周后,处死其中5只(空白对照组1只,AFL组4只)病理组织学检查以验证其造模成功,然后将AFL组进一步分为模型对照组、双葛解酲丸高剂量组、双葛解酲丸中剂量、双葛解酲丸低剂量组和海王金樽治疗组。治疗4周末处死所以大鼠,检测血清肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白介素-6(interleukin-6,IL-6)、痩素(leptin,LEP)、脂联素(adiponectin,ADP)和游离脂肪酸(free fatty acid,FFA)。同时对肝脏进行病理切片组织学检查。结果双葛解酲丸高剂量组大鼠血清IL-6[(62.49±34.16)pg/ml]、TNF-α[(3.25±0.20)ng/ml]、FFA[(572.16±45.37)μmol/L]和LEP[(1.89±0.21)ng/ml]明显低于模型对照组与海王金樽治疗组(P<0.01),血清ADP[(3.05±0.27)mg/ml]明显高于模型对照组[(2.17±0.37)mg/ml]与海王金樽治疗组[(2.69±0.32)mg/ml,P<0.01)],双葛解酲丸对大鼠肝脏的脂肪变性及炎症有明显改善。结论双葛解酲丸对酒精性脂肪肝有治疗作用。Objective To determine the efficiency of Shuangge Jiecheng Pill on the treatment of alcoholic fatty liver in the rat model.Methods A total of 65 SD rats were randomly divided into normal control group（n=11） and model group（n=54）.The rat model of alcoholic fatty liver disease was induced by Chinese liquor（56% Hongxing wine） by intragastric administration.One rat from normal control and 4 from the model group were used for model identification.The rest 50 rats of model group were further divided into Haiwang group（HW group,n=10,gastrically treated by Haiwang Jinzun Pill at 1.5 g/（kg·d）,twice per day for 4 weeks）,and high-,middle-and low-dosed Shuangge Jiecheng Pill groups.At the end of treatment,all rats were killed and their liver was taken for pathological changes.The serum levels of TNF-α,IL-6,leptin（LEP）,adiponectin（ADP） and free fatty acid（FFA） in the rats of all groups were measured and compared.Results High-dosed Shuangge Jiecheng Pill resulted in obvious decrease in serum levels of IL-6[（62.49±34.16）pg/ml],TNF-α [（3.25±0.20）ng/ml],FFA [（572.16±45.37）μmol/L] and LEP [（1.89±0.21）ng/ml] when compared with those of normal control and HW group（P0.01）,and significant increase in ADP [（3.05±0.27）mg/ml] compared with that of normal control[（2.17±0.37）mg/ml] and HW group[（2.69±0.32）mg/ml,P0.01].Pathological observation displayed obvious improvement in liver fatty changes and infliammation in Shuangge Jiecheng Pill treatment groups.Conclusion Shuangge Jiecheng Pill exerts curative effect on alcoholic fatty liver disease in rats.
