摘 要:目的探讨二次离心法制备富血小板血浆(PRP)的最佳离心条件和方法。方法从新西兰大白兔耳背中央动脉取血2.7 mL,按照不同离心条件(转速:1 000~2000 r/min,时间:5~20 min)离心2次,比较不同条件下PRP中血小板浓度和血小板回收率。结果多种离心条件均能成功制备PRP。初次离心:在形成白膜前的临界条件下(转速1 400 r/min,10 min)取得上层血浆的操作简单,且血小板回收率最高(〉85%)。再次离心:转速1 400 r/min、〉10 min时,即可制得符合要求的PRP(浓缩率〉5倍)。结论初次离心条件和上清液的获取方法对PRP的回收率和浓缩率影响较大,在产生白膜前的临界条件下提取上清液操作简单,无需特殊设备,血小板浓度和回收率高,避免了离心速度过高或离心时间过长导致的血小板活化、聚集。该方法是制备PRP简便、有效的方法。Objective To explore an ideal method for PRP concentration by double centrifugation.Methods Totally 2.7 milliliters whole blood was collected from central ear artery of the albino rabbit,and centrifuged according to different conditions(Speed:1 000-2 000 r/min;time:5-20 min).Calculating of the platelet concentration and recovery percent of platelet were performed.Results Several centrifuge conditions successfully prepared PRP.Initial centrifugation at 1 400 r/min for 10 min,which is the critical condition for the formation of Albuginea,achieved highest rate of platelet recovery,and the operation of upper plasma is simple.Second centrifuge condition was set for 1 400 r/min 10 min.Platelet counts in PRP prepared by four different methods were increased with respect to venous blood platelet counts.Percent recovery of platelet by the second method was observed to be significantly higher when compared to other methods.Conclusion Double centrifugation is an efficient method for PRP preparation.Initial centrifugation conditions at speed of 1 400 r/min for 10 min and followed with another centrifugation at 1 400 r/min( 10 minutes) has the higher platelet count and percent recovery of platelet.It could be an efficient method for PRP preparation.
