摘 要:目的进一步探讨硒对氟致雄性生殖损伤的干预作用。方法用氟化钠溶液和3个浓度梯度的亚硒酸钠溶液对雄性大鼠进行染毒,分为先硒后氟预防组系列和先氟后硒治疗组系列。对照组均用自来水代替硒。染毒完毕后测定大鼠睾丸组织中LDH、ACP、AKP、SOD、GSH-Px的活性和附睾中ATPase的活性。结果①与先水后氟对照组相比:低硒后氟组LDH、AKP、SOD活性显著升高(P<0.05);中硒后氟组GSH-Px活性极显著升高(P<0.01);3个先硒后氟组ACP活性均极显著升高(P<0.01)。低硒后氟组和中硒后氟组Ca2+-ATPase活性极显著(P<0.01)和显著升高(P<0.05)。②与先氟后水对照组相比:氟后低硒组LDH活性极显著升高(P<0.01);氟后中硒组Na+,K+-ATPase、Mg2+-ATPase活性显著升高(P<0.05),Ca2+-ATPase活性极显著升高(P<0.01)。结论硒对氟致大鼠生殖损伤的干预方式中,预防效果好于治疗效果,其中0.375 mg/L是本实验条件下的最佳预防作用剂量,LDH可能是预防作用的特异性药物靶点。Objective To further explore the interference of selenium on male reproductive toxicity induced by excessive fluorine in male rats.Methods Prevention group: The rats in fluoride-after-water control group drank tap water before the administration of 50 mg/L of sodium fluoride solution.Three fluoride-after-selenium groups were asked to drink three kinds of sodium selenite solution,then drank 50 mg/L sodium fluoride solution.Treatment group: The feeding sequence was opposite to the prevention group.Results The enzyme activities of LDH,ACP,AKP,SOD,GSH-Px,and ATPase were examined.The results are as follows ①prevention group: compared with the control the activitives of LDH、AKP、SOD were significantly(P0.05) increased in fluoride-after-low-dose selenium group while the activity of GSH-Px was significantly(P0.01) increased in fluoride-after-mid-dose selenium;Similar significant(P0.01) increased was found in the activity of ACP in all the fluoride-after-selenium groups respectively.The activity of Ca2+-ATPase in the fluoride-after-mid-dose selenium and fluoride-after-low-dose selenium had significantly(P0.05) and more significantly(P0.01) increased.②Treatment group: Compared with the control group,the activity of LDH were significantly(P0.01) increased in low-dose selenium-after-fluoride group.Significant increase(P0.05) in the activity of Na+K+-ATPase was observed in the mid-selenium group;while the activity of Ca2+-ATPase increased more significantly(P0.01).Conclusion The prophylactic effect of selenium is better than therapeutic effect according to this experimental conditions.Na2SeO3 sodium selenite solution of 0.375 mg/L is the optimal dose for prevention against fluorosis on reproductive damage.LDH is likely to be a specific drug target of the selenium as a prevention on fluorosis.
