摘  要:目的观察苦参碱(matrine,Ma),对受低剂量照射小鼠骨髓细胞端粒酶活性及细胞凋亡的影响。方法采用Varine600直线加速器为照射源,以0.2Gy/d的小剂量分次照射小鼠,分别于照射5、10、15、20天后的1、3、6、10、15、21天分批活杀小鼠,涂片观察小鼠骨髓细胞的增生程度,采用端粒重复扩增(telomeric repeat amplification protocol,TRAP)-酶联免疫吸附剂测定(enzyme-linked immunosorbent assay,ELISA)照射后小鼠骨髓细胞的端粒酶活性;采用AnnexinV-FITC及PI双染、应用FACScan进行流式细胞术定量检测细胞凋亡率,观察未服用苦参碱、照射后不同累积照射剂量及服用不同剂量苦参碱后不同时间的小鼠骨髓细胞端粒酶活性、细胞凋亡率的改变。结果低剂量分次照射小鼠10天内小鼠骨髓增生明显,骨髓细胞端粒活性高表达,细胞凋亡减少(P〈0.05);当照射大于10天时骨髓增生明显受抑,抑制端粒酶活性表达,诱导细胞凋亡(P〈0.05);高剂量苦参碱服用10~20天时可显著抑制小鼠骨髓细胞端粒酶活性高表达及诱导细胞凋亡(P〈0.05)。结论低剂量分次照射小鼠(累积剂量〈2Gy)可诱导骨髓细胞端粒酶活性高表达,并抑制细胞凋亡;累积剂量〉2Gy则产生相反作用。高剂量苦参碱可抑制小鼠骨髓细胞端粒酶活性高表达,诱导骨髓细胞凋亡,可作为辐射防护的一种选择。Objective To observe the effect of matrine on telomerase activity and apoptosis of bone marrow cells in mice exposed to low-dose radiation.Methods Mice were exposed 0.2 Gy/d irradiation by Varine 600 linear accelerator for 5 days,10 days,15 days and 20 days respectively,and sacrificed on the 1st,3rd,6th,10th,15th and 21st day after irradiation respectively.The extent of bone marrow cell proliferation were observed.Telomerase activity of the bone marrow cells was detected by telomerie repeat amplification protocol-enzyme-linked immunosorbent assay(TRAP-ELISA).The apoptosis rate was detected by flow cytometry(FCM) after marked by Annexin V-FITC and PI.Results The proliferation of bone marrow cells was obviously increased in mice within 10 days after irradiation,the telomerase activity was highly expressed,and the apoptosis was decreased(P〈0.05).Ten days after low-dose irradiation,the proliferation and telomerase activity were inhibited and the cell apoptosis was induced(P〈0.05).High dose matrine administration could significantly inhibit the high expression of telomerase and induce the cell apoptosis ten to twenty days after irradiation(P〈0.05).Conclusion Low-dose irradiation with the accumulative dose lower than 2 Gy can induce the high expression of mice bone marrow cell telomerase activity and inhibit the cell apoptosis.The accumulative dose higher than 2 Gy has the opposite effect.High dose matrine can inhibit the high expression of mice bone marrow cell telomerase activity,induce the cell apoptosis,and can be chosed for radioprotection.
