椰乳和香蕉泥对东方百合杂交幼胚离体培养的影响

摘　　要：探讨椰乳和香蕉泥、胚龄以及杂交组合对百合幼胚离体培养的影响。以东方百合的6个杂交组合、3个胚龄的幼胚为试材进行离体培养试验。试验结果表明:胚龄60～65d的幼胚离体培养成活率最高,其次为80～85d的幼胚,再次为70～75d的幼胚。不同杂交组合的幼胚离体培养的平均成活率从高到低依次为:罗蒂娜×马可波罗,罗蒂娜×法兰西,罗蒂娜×索蚌,提拔×法兰西,提拔×康斯坦萨,提拔×马可波罗。在60～65d胚龄时,在培养基中加入50mg.L-1椰乳使幼胚的成活率提高8%,加入100mg.L-1椰乳可使成活率提高5%,但加入香蕉泥使成活率大幅降低;在70～75d胚龄时,加入香蕉泥效果稍好;在80～85d胚龄时,加入椰乳和香蕉泥的培养基效果都较差。不同胚龄的幼胚离体培养成活率存在极显著差异,刚刚固体化的幼胚成活率最高。杂交组合不同幼胚离体培养成活率不同。培养基中添加适量椰乳能提高刚刚固化的幼胚的成活率,添加适量的香蕉泥能提高固化程度稍高的幼胚的成活率,对发育到胚乳完全硬化程度的百合胚,加入椰乳和香蕉泥均有不利影响。The purpose of this article is to explore the influence of coconut milk,mashed bananas,embryo age and crosses to lilium young embryo culture in vitro.with oriental hybrids six crosses,three embryos of embryonic age as materials for in vitro test.The results showed that there were the highest survival rate from the embryo of 60 ～ 65 days,followed by 80～ 85d,the worst 70～75d.The average survival rate of embryo culture in vitro is in the order,Rodina× Marco Polo,Rodina ×Francia,Rodina × Sorbonne,Tiber×Francia,Tiber× Constanta,Tiber × Marco Polo.Medium supplemented with natural extracts had significant effect on embryo survial rates.The survival rate of 60～65 days young embryo was increased 8% by supplemented with 50 mg·L-1 coconut milk at the medium,adding 100mg·L-1 increased by 5%,but adding mashed banana to significantly lower survival rate.The 70～75d embryos increased slightly in the medium adding mashed banana.Adding coconut milk and mashed banana medium effects are worse in 80-85d.Different age young embryo in vitro culture survival rate existed very significant difference and just the solidification young embryo survival rate was the highest.The survival rate of embryo culture was different from different hybrid combinations.Medium add appropriate coconut milk improved just cured young embryo survival rate.The survival rate of the harder embryo could be increased in the medium adding proper mashed bananas.The survival rate of the lily embryos from hardened endosperm was lower in the medium added coconut milk and mashed bananas.
