健脾、补肾和健脾补肾三法对慢性乙型肝炎患者外周血树突状细胞功能的影响

摘　　要：[目的]比较健脾法、补肾法和健脾补肾法对慢性乙型肝炎(CHB)患者外周血树突状细胞(DCs)功能的影响。[方法]收集24例CHB患者和10例健康人的抗凝外周静脉血,分离外周血单个核细胞(PBMCs),进一步分离提取培养DCs,采用大鼠健脾、补肾和健脾补肾药物血浆进行干预。检测DCs细胞表面分子CD1α、CD80、CD86及藻红蛋白(HLA-DR)分子的表达。[结果]CHB组DCs表面分子CD1α、CD80、CD86及HLA-DR的表达较健康对照组低(P<0.05)。3组药物血浆干预后上述DCs表面分子表达率均有升高。其中,补肾药物血浆干预后DCs表面分子CDlα、CD80、CD86、HLA-DR表达率较干预前均有明显提高(P<0.05),且CD1α、CD86及HLA-DR表达率的提高较另2组明显(P<0.05)。[结论]健脾法、补肾法和健脾补肾法均能调节CHB患者DCs细胞免疫功能,以补肾法影响更为明显。[Objective]To investigate the effect of different treatments of TCM（tonifying kidney, strengthening spleen and tonifying kidney combined with strengthening spleen）on DC cells in patients with chronic hepatitis B（CHB）. [Methods]Twenty-four patients with chronic hepatitis and ten healthy controls were included in this study. DC cells of phenotype CDIα, CDSO, CD86 and HLA-DR were differentiated from peripheral blood mononuclear cells （Pt3MCs）, incubated and induced. Then the DC cells were interfered by plasma of medicine of different treatments of TCM. The expressions of CDIα, CD80, CD86 and HLA-DR cells were detected by FACS.[Re sults]The levels of CDIα, CD80, CD86 and HLA-DR were much lower in CHFI patients than in healthy controls（P〈0.05）. After interfered by plasma of medicine of three treatments of TCM, the levels of DC cell phenotypes were increased. The levels of DC cell phenotypes were much higher in tonifying kidney group （P〈0.05）, and the levels of CDIα, CD86 and HLA-DR were higher in tonifying kidney group than other grout）s （P〈0. 05）. [Conclusion]The three trentmentn of TCM can regulate DC cells in patients with chronic hepatitis. The influence of tonifying kidney on DC cell function in patients with chronic hepatitis is more prominent.
