摘 要:目的:建立南沙参中多糖含量的测定方法并进行比较。方法:采用水提醇沉法分离提取南沙参多糖,以葡萄糖为对照制备标准曲线;采用蒽酮-硫酸法和苯酚-硫酸法,利用紫外分光光度计测定多糖含量。结果:蒽酮-硫酸法测定南沙参中多糖含量为16.37%,加样回收率为101.1%,RSD为2.1%;苯酚-硫酸法测定南沙参中多糖含量为15.26%,加样回收率为99.9%,RSD为1.8%。结论:两种方法操作简单,精密度高、重复性好,均可用于南沙参中多糖含量的测定。Objective:To establish and compare the methods for the content determination of polysaccharides in Radix Adenophorae. Method:The polysaccharides were isolated and extracted from Radix Adenophorae by water extraction and alcohol precipitation. The standard curve of polysaccharides from Radix Adenophorae was obtained with glucose as the control.The content of polysaccharides was determined by anthrone sulphuric acid colorimetry and phenol-vitriolic colorimetry using a UV spectrophotometer.Result:The content determined by anthrone-sulphuric acid colorimetry was 16.37%with average recovery of 101.1%(RSD =2.1%),and that determined by phenol-vitriolic colorimetry was 15.26%with average recovery of 99.9%(RSD = 1.8%).Conclusion:Both methods for the content determination of polysaccharides in Radix Adenophorae are reliable and accurate,and can be considered as the assay methods for polysaccharides in Adenophora tetraphylla(thunb.) Fisch.
