柯萨奇B1型病毒的鉴定及VP1区基因特征分析

摘　　要：目的分离引起手足口病的肠道病毒,分析其基因序列特征,为手足口病的防控提供科学依据。方法从手足口病病例标本中分离病毒,提取病毒核酸并特异性扩增VPl区基因片段,进行序列测定、系统发生树构建和核苷酸同源性分析。结果从2009年河南省报告的1例手足口病病例标本中分离出1株柯萨奇B1型肠道病毒,基因序列显示其与浙江省2009年分离株同源性最高,核酸同源性达到98.4%,氨基酸同源性达100%。结论河南省手足口病病原谱中存在柯萨奇B1型病毒,并与浙江分离株属同一分支。Objective To isolate and identify an enterovirus strain isolated from a patient of hand foot and mouth disease （HFMD）. Methods Enterovirus was isolated in 2009 using chloroform and glass beads shaking, supernatant was inoculated to Hep-2 ceils. A fragment spanning the VP1 region of the coxsackievir- us was amplified using RT-PCR and sequenced for genotyping analysis. Results An enterovirus strain was isolated from clinical specimen. Sequencing analysis of the coxachievirus VP1 region classified the isolate into coxsackievirus B1 （CVB1）, this strain shares the highest nucleotide and amino acid homology （98.4o/00 and 100%, respectively） with the strain isolated in Zhejiang province in region VP1. Conclusions Coxsackie- virus B1 isolated from a HFMD patient in Henan province belongs to the same family with the stain identified in Zhejiang.
