摘 要:目的观察吡那地尔诱导失血性休克大鼠血流动力学的保护及与线粒体ATP激活钾通道(K栅)的关系。方法采用失血性休克复苏大鼠,观察吡那地尔预处理对大鼠存活时间、血流动力学指标[平均动脉压(MAP)、左室收缩压(LVSP)、左室舒张末压(LVEDP)、左室压最大上升/下降速率(4-ap/dtmax)、心率(HR)]的影响,以及线粒体KATP关闭剂5-羟基癸酸(5-HD)和格列本脲对其效应的作用。结果吡那地尔预处理显著延长大鼠存活时间,恢复血流动力学,MAP、LVSP、LVEDP、+do/dtmax、-dp/dtmax、HR分别增高5.9%、11.6%、32.9%、28.0%、28.1%和13.4%(P〈0.01),5-HD和格列本脲可显著抑制其保护效应(P〈0.01)。结论吡那地尔开放线粒体KATP,保护失血性休克大鼠血流动力学,提高存活率。Objective To observe the pinacidil pretreatment-induced protective effects on the he- modynamic of hemorrhagic shock rats, and the mediated role of mitochondrial KATe channel. Methods U- sing hemorrhagic shock rats, the animal survival time and the hemodynamic parameters including mean ar- terial blood pressure (MAP), left intraventricular systolic pressure (LVSP), left ventricular end-diastolic pressure (LVEDP) , maximal change rate of left intraventrieular pressure ( + dp/dtmax) and heart rate (HR) were observed after pinacidil pretreatment and shock-resuscitation, and the effects of mitochondrial KATe channel blocker 5-hydroxydecanoate (5-HD) and glibenclamide on pinacidil pretreatment-induced a- bove protection were observed to interpret its possible mechanism. Results As compared with simple fluid resuscitation, combination of fluid resuscitation with pinaeidil pretreatment significantly prolonged the sur- vival time of shock-resuscitation rats and increased the hemodynamic parameters (MAP, LVSP, LVEDP, +dp/dtmax, - dp/dtmax and HR were increased by 5.9% , 11.6%, 32.9%, 28.0% , 28. I% and 13.4%, respectively (P 〈0. 01 ), and all these effects could be significantly blunted by 5-HD and glib-enelamide (P 〈 0.01 ). Conclusion Pinacidil pretreatment induced the protective effects on the hemody-namic of hemorrhagic shock rats and prolonged the animal survival time through mitochondrial KATp chan- nel.
