摘 要:羊毛角蛋白的溶解已经有几十年的历史,但一直没有发现较好的溶解方法。首次将生化用新型精细二硫键还原剂TCEP(三羧乙基膦)应用于羊毛角蛋白的溶解。TCEP对二硫键有很强的选择还原性,与亚硫酸氢钠联合使用,与以往的羊毛溶解方法相比有很大优势,使羊毛的溶解效率大大提高,溶解时间为5h时,溶解率达96.3%。通过红外测试推断,少量的不溶物为胱氨酸含量较高的高或超高硫蛋白。角蛋白浓度达10%,所制得的角蛋白溶液分子量在3500D以上的达80%,8000D以上的达72%。通过凝胶电泳分析,得到角蛋白的主体分子量为14000~20000D。The dissolution of the wool keratin has a history of several decades, but has not found an effective dissolution method. A novel biochemical disulfide reducing agent-TCEP(Tris(2-carboxyethyl) phosphine) was applied to the dissolution of wool keratin for the first time. TCEP has a strong selection of disulfide reduction, and joint use of sodium bisulfite, the dissolution of the previous methods have great advantages in wool. The efficiency of dissolution was greatly improved; dissolution time was 5h, dissolution rate was 96.3%. Through FT-IR, the small amount of in- soluble matter was high or ultra high sulfur protein which has a high content of eystine maybe. Keratin concentration was 10%, the molecular yield of keratin more than 3500D and more than 8000D were 80% and 72%. By SDS-PAGE, the body molecular of the keratin was 14000-20000D.
