摘 要:目的:观察木丹颗粒对糖尿病大鼠胰岛细胞凋亡的作用及TNF-α、caspase-3表达影响。方法:利用链脲佐菌素腹腔注射法诱导胰岛细胞凋亡。将实验用SD大鼠随机分正常对照组(NC)、DM组、木丹颗粒低、中、高剂量组、甘精胰岛素组,给药4周。于给药后2周、4周测定大鼠空腹血糖(FPG),TUNEL法检测胰岛细胞凋亡,免疫组化测定TNF-α、Caspase-3的表达。结果:①与NC组比较,DM组大鼠空腹血糖明显升高(P<0.01),胰岛细胞凋亡显著增多,TNF-α、Caspase-3表达强阳性。②与DM组比较,木丹颗粒中高剂量组空腹血糖无显著下降(P>0.05),甘精胰岛素组空腹血糖明显下降(P<0.01)。木丹颗粒的中高剂量可下调TNF-α、Caspase-3的表达。结论:木丹颗粒无显著的降糖作用,但可降低胰岛细胞内TNF-α、Caspase-3的表达,减少凋亡,从而对胰岛细胞具有保护作用。Objetive : To investigate the effect of the Mudan granule against DM rats by observing the changes of TNF- α and caspase-3 protein in pancreatic beta cells. Methods : Islet cell apoptosis was induced by intraperitoneal injection of strepotozoein ( STZ ) .The rats were randonmLy allocated into normal control group ( NC group ), diabetes mellitus group ( DM group ), Mudan granule low dosage group, Mndan granule middle dosage group, Mudan granule high dosage group, Insulin Glargine Injection group, all treated 4weeks. During the 2nd week and after the treatment, fasting plasma glucose ( FPG ), expressions of TNF- ot and caspases-3 in rat islet cells were measured by immunohisto chemistry, and the cell apoptosis was analyzed by TUNEL assay. Results : The rats in the DM group showed FPG was higher in model group than that of the normal group (P〈0.01), Mudan granule significantly increased expressions of TNF-oL and caspases-3, and reduced the islet cell apoptosis of the diabetic rats; Mudan granule could decrease FPG ( P〉0.05 ), and significantly inhibit expressions of TNF- α and caspase-3 in rat islet cells. Insulin Glargine Injection treatment significantly decreased FPG (P〈0.O1).Conclusion: Mudan granule can decrease expressions of TNF- α , caspase-3 and inhibit the apoptosis of beta cells in islet, which may be one of the mechanisms of Mudan granule improving DM.
