摘  要:目的:研究26-nor-8-羟基-α-芒柄花醇(26-nor-8-hydroxy-α-onocerin,26HO)对成骨细胞活性的影响。方法:采用MTT法研究不同浓度的26HO对成骨细胞增殖率的影响;并用Real-Time PCR研究不同浓度的26HO对成骨细胞成骨相关基因表达量的影响。结果:20μM/L和40μM/L的26HO短期处理(1d)能够促进成骨细胞增殖,长期处理对成骨细胞增殖活性没有显著影响;26HO对骨相关基因(ALP、OCN、Col-I)表达的影响与成骨细胞的成熟状态有关。而对AP-1转录因子家族基因中Fos亚家族成员(c-fos、Fra-1、Fra-2)等基因mRNA表达的影响与26HO浓度有关,与处理时间无关;而对AP-1转录因子家族基因中Jun亚家族成员(c-Jun和Jun-D)基因mRNA表达的影响与26HO浓度无关,与处理时间有关。结论:26HO具有成骨活性。Objective:To investigate the effects of 26-nor-8-hydroxy-α-onocerin(26HO) on osteoblast activity.Methods:MTT assay was used to analyze the effect of 26HO on osteoblast proliferation and Real-Time PCR was applied to explore the effect of 26HO on the expression of osteogenic genes in osteoblasts.Results:One-day treatment of 20μM/Land 40μM/L 26HO significantly promoted osteoblast proliferation,but long-term intervention showed no obvious effect on osteoblast proliferation.The effect of 26HO on bone-related gene expression was related to the maturation status of osteoblast being treated.The effect of 26HO on the expression of fos sub-family of AP-1 transcription factor family is related to concentration of 26HO but not treatment period,while the effect of 26HO on the expression of Jun sub-family of AP-1 transcription factor family was related to treatment period but not concentration of 26HO.Conclusion:26HO has osteogenic activity.
