摘 要:目的探讨益气养阴消癥通络中药对高糖联合血管紧张素Ⅱ(AngⅡ)培养的大鼠肾小球系膜细胞分泌转化生长因子β1(TGF-β1)、结缔组织生长因子(CTGF)的影响,为进一步研究其防治糖尿病肾病(DN)的作用机理提供依据。方法原代培养肾小球系膜细胞(MCs),实验共分为5组:低糖对照组、高糖组、高糖+AngⅡ组、中药血清组、厄贝沙坦血清组。于第5代时,予高糖、高糖联合AngⅡ刺激,并同时进行益气养阴消癓通络中药及厄贝沙坦西药血清干预,48 h后ELISA法检测细胞上清TGF-β1、纤维连接蛋白(FN)、层粘连蛋白(LN)的蛋白含量,Western blot检测CTGF蛋白表达水平。结果与低糖对照组比较,其他组TGF-β1、CTGF、FN、LN表达均明显升高;与高糖+AngII组比较,厄贝沙坦血清、中药血清组TGF-β1、CTGF、FN、LN表达均明显下降(P<0.01)。结论益气养阴消癥通络中药可能通过抑制CTGF表达而有效降低TGF-β1的水平,从而抑制细胞外基质的合成,发挥其保护肾脏的作用。Objective To investigate the influences of Chinese medicinal with actions of replenishing qi and nourishing yin,and resolving mass and freeing collateral vessels on transforming growth factor β1(TGF-β1) and connective tissue growth factor(CTGF) secreted by mesangial cells(MC) in rats fostered by high glucose combining Ang II,and provide a foundation for further study on prevention and treatment of diabetic nephropathy(DN).Methods The fifth generation of primary cultured MC was stimulated with high glucose and high glucose combing Ang II as well as Chinese medicinal serum.After 48 hours,the levels of TGF-β1,fibronectin(FN) and laminin(LN) were detected by using ELISA,and expression of CTGF was detected by using Western blotting method.Results Compared with control group,the levels of TGF-β1,FN and LN and CTGF expression increased significantly in other groups.Compared with high glucose+AngII group,the levels of TGF-β1,FN and LN and CTGF expression decreased significantly in irbesartan group,Chinese medicinal serum group(P0.01).Conclusion Chinese medicinal with actions of replenishing qi and nourishing yin,and resolving mass and freeing collateral vessels can effectively decrease the level of TGF-β1,inhibit the composition of extracellular matrix and protect kidney through inhibiting CTGF expression and reducing TGF-β1 level.
