摘 要:目的优选大孔吸附树脂分离纯化蜘蛛香中环烯醚萜类成分。方法采用高效液相色谱法测定蜘蛛香中环烯醚萜类成分11-ethoxyviburtinal的含量,并以此为指标,采用单因素考察、正交试验设计法对上样溶液浓度、洗脱溶剂、洗脱流速等进行考察及优化。结果 AB-8大孔吸附树脂具有良好的吸附分离性能。确定工艺参数为:上样浓度为0.2 g/mL,树脂柱径高比为1∶6,除杂溶剂为2倍柱体积(BV)的30%乙醇,洗脱溶剂为90%乙醇,洗脱体积为5 BV,洗脱流速为4 BV/h。结论此法操作方便,为大生产提取分离纯化蜘蛛香提供了依据。Objective To optimize macroporous resin for isolating and purifying the iridoid constituents from Zhizhuxiang(Jatamans Valeriana Rhizome).Methods The content of 11-ethoxyviburtinal was determined by using HPLC and then taking which as index,the sample solution concentration,elution solvent and elution flow rate were studied and optimized by applying single factor observation and orthogonal test design.Results AB-8 macroporous resin had excellent adsorption and desorption properties.The technological parameters were determined as that sample concentration was 0.2 g(crude Zhizhuxiang)/mL,ratio of column diameter to height was 1:6,purification solvent was 30% ethanol(2 BV),elution solvent was 90% ethanol,elution volume was 5 BV and elution flow rate was 4 BV/h.Conclusion The method was easy in operation,which provides a foundation for isolating and purifying Zhizhuxiang in large scale production.
