摘 要:目的:测定3种方法(传统单剂煎煮法、煎药机多剂煎煮法、配方颗粒溶解法)制备的胃肠复原汤中黄芪甲苷的含量并进行比较。方法:采用高效液相色谱法。色谱柱为KromasilC18(150mm×4.6mm,5μm),流动相为乙腈-水(31:69),流速为1.0mL.min-1,检测器为Alltech2000ES蒸发光散射检测器,漂移管温度为105℃,气体流速为2.0mL.min-1。结果:黄芪甲苷进样量在0.4832~1.4496μg范围内与峰面积积分值呈良好线性关系(r=0.9993);平均加样回收率为97.21%,RSD=1.45%(n=6)。胃肠复原汤3批传统煎剂平均含量为0.2250mg.g-1,3批机制煎剂平均含量为0.1545mg.g-1,3批配方颗粒平均含量为0.2742mg.g-1。结论:胃肠复原汤中黄芪甲苷含量配方颗粒稍高于传统煎剂,二者均远远高于机制煎剂。批间差异配方颗粒与传统煎剂接近,而机制煎剂差异较大。OBJECTIVE:To determine the content of astragaloside Ⅳ in Weichang fuyuan decoction prepared by 3 kinds of methods(traditional single dose decoction method,decoction machine multiple decoction method,magistral granule dissolution method).METHODS:HPLC method was adopted.The determination was performed on Kromasil C18(150 mm×4.6 mm,5 μm)column with mobile phase consisted of acetonitrile-water(31:69) at the flow rate of 1.0 mL·min-1.Alltech 2000ES evaporative light scattering detector was adopted with drift tube of 105 ℃ and flow rate of 2.0 mL·min-1.RESULTS:The linear range of astragaloside Ⅳ was 0.483 2~1.449 6 μg(r=0.999 3) with an average recovery of 97.21%(RSD=1.45%,n=6).The average content of 3 batches of traditional Weichang fuyuan decoction was 0.225 0 mg·g-1,that of 3 batches of decoction prepared by machine was 0.154 5 mg·g-1 and that of 3 batches of magistral granule was 0.274 2 mg·g-1.CONCLUSION:The content of astragaloside Ⅳ in Weichang fuyuan decoction of magistral granule is slightly higher than traditional decoction,and they are much higher than decoction prepared by machine.The difference of batches of magistral granule is similar to that of traditional decoction,but decoction prepared by machine has great difference.
