摘  要:目的为探讨人类呼吸道腺病毒感染自限性的分子机制,对人类常见呼吸道腺病毒5型(AdS)和7型(Ad7)感染对呼吸道上皮细胞抑炎分子——黏蛋白1(MUC1)表达的影响进行初步研究,并探讨两者差异性。方法分别用Ad7和AdS感染呼吸道上皮细胞A549构建感染模型。qRT-PCR和Westernblot分别检测感染后MUC1mRNA转录水平及蛋白表达水平变化。结果Ad5感染A549细胞后MUCImRNA转录水平及MUC1蛋白表达水平均可见上调,且呈一定的时间效应;而Ad7感染后未观察到此现象,延长Ad7感染时间后,仍未观察到MUC1mRNA转录上调。结论人类呼吸道AdS感染呼吸道上皮细胞初期,可诱导上调抑炎分子MUC1表达,提示MUC1全部或部分参与到Ad5感染的自限性过程。但Ad7感染不能诱导上调MUC1表达,其感染的自限性来源于其他机制。Objective To explore the molecular mechanism for the self-limitation of adenoviral in- fections in human airway, the different impacts of adenovirus serotype 5 ( AdS ) and serotype 7 ( Ad7 ) infections on ~nucin 1 ( MUC1 ) expression in airway epithelial cells were preliminarily investigated. Methods The AdS and the Ad7 infection models were established in A549 cell line. qRT-PCR was performed to deter- mine the transcription of MUCI mRNA, and the expression of MUC1 in A549 cells infected by AdS or Ad7 was by detected Western blot. Results An up-regulation of the MUC1 mRNA level were observed after AdS infection for 6 h(P〈0.05 ), and the protein expression level of MUC1 increased in a time-dependent manner in 48 hours of AdS infection , while similar response of MUCI mRNA was absent in Ad7 infection (6 h), even after prolonged ( 20 h ) treatment ( P 〉 0.05 ). Conclusion This study reveals an up-regulation of MUCI expression as one of the early immune response to AdS infection, which implies that MUC1 may func- tion fully or partially as an anti-inflammatory factor in the self-limitation effect of AdS infection. However, type7 adenoviral infection, may introduce a mechanism otherwise, but through MUC1.
