摘 要:目的研究三白草中主要化学成分对H2O2损伤人正常肝细胞LO2的保护作用。方法供试品为三白草中木脂素类成分:Rel-(7S,8R,7’R,8’R)-3,3’,4,4’,5,5’-hexamethoxy-7-O-7’,8,8’-llignan、Di-O-methyltetra hydrofuriguaiacin B、里卡灵A、里卡灵B、三白草素、二氢愈创木酸;黄酮类成分:异槲皮苷、槲皮苷。LO2细胞接种于96孔板培养,加入100μmol/L的受试成分,培养24h后加入20μL 1.2mmol/L H2O2作用1h。使用MTT法检测各组细胞存活率。结果异槲皮苷和槲皮苷对H2O2损伤LO2细胞有显著保护作用,里卡灵A和二氢愈创木酸对细胞有显著损伤作用。结论该实验可以为三白草保肝作用的研究提供理论基础。OBJECTIVE To study the protective effect of major contents from saururus chinensis on H2O2-induced LO2 cells damage.METHODS Tested samples are lignanoids: Rel-(7S,8R,7'R,8'R)-3,3',4,4',5,5'-hexamethoxy-7-O-7',8,8'-llignan、Di-O-methyltetra hydrofuriguaiacin B、Licarin A、Licarin B、Saucernetin and Dihydroguaiaretic acid;flavonoids : Isoquercitrin and Quercitrin.LO2 cells were cultured in 96 well plates.Then 100 μmol/L of tested samples were added.After 24 h,cells were treated with 1.2 mmol/L H2O2 for 4 h.MTT assay was used to detect survival rate of LO2 cells.RESULTS Isoquercitrin and quercitrin had distinguished protective effect,but licarin A and dihydroguaiaretic acid had distinguished damaging effect.CONCLUSION The study can be used as research theoretic basis of liver protection from saururus chinensis.
