反相高效液相色谱法测定茶碱缓释片中茶碱的含量

摘　　要：目的建立茶碱缓释片中荼碱的含量测定方法。方法采用反相高效液相色谱法，HypersilCl8柱（5μm，150mm×4．6mm），以甲醇-水（30：70）为流动相，流速为1．0ml／min；检测波长203nm；柱温为室温，进样量为20μl。结果茶碱在4-36ug／m1浓度范围内线性关系良好，r=0．9999；平均回收率为99．2％，方法精密度（RSD）为0．29％（n=5）。结论反相高效液相色谱法可用于茶碱缓释片中茶碱的含量测定，以控制茶碱缓释片的质量。Objective To establish a reversed-phased high performance liquid chromatogra- phy （RP-HPLC） method for the determination of Theophylline sustained release tablets.Methods By using RP-HPLC, a Hypersil C18 column was used （5μ m, 150 mm× 4.6 mm）, with the mobile phase of methanol-water（30 ： 70）, the flowing rate was 1.0 ml/min, and the detection wavelength was 203 nm in the room temperature.Results Theophylline was linear over the range of d - 36 μ g/ml（r=0.999 9, n=5）.The average recovery was 99.2% （RSD=0.29%, n=5）.Conclusion It＇s a feasible method for the detemaination of theophylline so as to control the quality of theophylline sustained release tablets.
