阳痿肾阳虚证转录组学特征研究

摘　　要：目的:研究阳痿肾阳虚证的转录组特征,揭示肾阳虚证"同证异治"的生物学基础。方法:分别对3例阳痿肾阳虚证,3例健康组进行Agilent人4×44 k表达谱芯片实验,对差异表达基因进行GO、Pathway分析。结果:阳痿组与健康组的差异表达基因达1396条。其中差异性最大的是归属于GO:0008287(蛋白丝氨酸/苏氨酸磷酸酶复合物),找出具有显著性差异的信号转导通路8个。结论:阳痿肾阳虚证的转录组特征有可能是丝氨酸/苏氨酸磷酸酶复合物通过Rho激酶调节钙信号转导通路,通过与前期同类研究对比,发现阳痿肾阳虚证在钙信号转导通路方面存在一致性,其他功能基因及通路均有差异。由此表明不同疾病的肾阳虚证在信号转导通路方面各自有显著特点,为我们提出的"同证异治"提供了分子生物学基础上的证据。Objective： To study the Kidney Yang deficiency syndrome of erectile dysfunction from the transcription of the characteristics of group,and reveals the kidney Yang deficiency syndrome ＂ treating same pattern with different method ＂ the biological basis.Methods： 3 cases of Kidney Yang deficiency syndrome of erectile dysfunction,3 cases health groups Agilent people 4×44 k expression profile chip experiment,differentially expressed genes GO,Pathway analysis.Results： the group and the group health erectile dysfunction differentially expressed genes article 1396.One of the biggest differences is relegated to GO： 0008287（protein serine/threonine phosphatase complex）,find out has significant difference between the signal transduction pathways of eight.Conclusion： Kidney Yang deficiency syndrome of erectile dysfunction of transcription group characteristics may be serine/threonine phosphatase Rho kinase complex through regulating calcium signal transduction pathways through the contrast with similar research,found the kidney Yang deficiency syndrome in calcium signalling pathways are consistent,and other functional genes and pathways are different.This suggests that different disease of kidney Yang has its own marked characteristics,accord with our early research suggests kidney Yang deficiency syndrome because different diseases,there are many fine syndrome.It also presented by the ＂ treating same pattern with different method ＂ provide the molecular biology on the basis of evidence.
