摘 要:目的观察榛花总黄酮对缺血性肾病大鼠肝脏丙二醛(MDA)、超氧化物歧化酶(SOD)、一氧化氮(NO)含量的影响及对慢性缺血性肾病的治疗作用。方法将90只SD大鼠随机分为治疗组(予榛花总黄酮)、对照组(予生理盐水)和假手术组。在治疗组和对照组中复制大鼠缺血性肾病模型,在第14,28,45天分批处死大鼠,采用酶联免疫吸附法检测缺氧诱导因子(HIF-1α)、结缔组织生长因子(CTGF)、Ⅰ型胶原蛋白的水平,在第28天的大鼠肝脏中检测NO、MDA及SOD的含量。结果榛花总黄酮可明显降低HIF-1α,CTGF、Ⅰ型胶原在缺血性肾病中的表达,也可使SOD活性和NO含量明显升高,MDA含量明显降低。结论榛花总黄酮可通过减少缺血肾病中氧自由基和HIF-1α的含量,抗脂质过氧化,减少纤维化因子的表达和肾间质纤维化的形成。Objective It is to observe the effect of total flavones from Hazel's flower on the contents of MDA,SOD,NO in liver of rats with ischemic nephropathy,to explore the therapeutic effect of this drug on the disease.Methods 90 rats were randomly divided into 3 groups: treatment group(total flavones from Hazel's flower),control group(normal saline) and sham group.The renal ischemic nephropathy model was established in the treatment group and control group.The rats were killed in 14,28,45 days in batches.The protein expression of hypoxic inducible factor-1α(HIF-1α),connective tissue growth factor(CTGF),I type collagen in rat liver were detected by ELISA.The contents of nitric oxide(NO),malondialdehyde(MDA) and superoxide dismutase(SOD) were detected in rat liver in the 28 days' batch.Results Total flavones from Hazel's flower reduced the expression of HIF-1α,CTGF,I-type collagen in ischemic nephropathy rats models,significantly increased SOD activity and NO(P0.05),and also significantly decreased MDA(P0.05).Conclusion Total flavones from Hazel's flower have the effect of anti-lipid peroxidation and reduce fibrosis factor and renal interstitial fibrosis by reducing oxygen free radicals and HIF-1α.
