摘 要:目的研究中药有效成分小檗碱、苦参碱抗外阴阴道念珠菌病混合菌生物被膜形成的作用。方法体外建立白念珠菌、铜绿假单胞菌混合菌的生物被膜,采用四唑鎓盐(tetrazolium salt,XTT)减低法及形态学方法观察白念珠菌混合菌生物被膜的形成过程;采用形态学观察、活菌计数法评价小檗碱、苦参碱对白念珠菌混合菌生物被膜的最小抑制浓度。结果白念珠菌混合菌培养48h形成成熟的生物被膜;125mg/L小檗碱能抑制白念珠菌混合菌生物被膜的形成。500mg/L苦参碱未见有抑制白念珠菌混合菌生物被膜形成的作用。结论小檗碱具有抗生物被膜形成的作用,可用于预防外阴阴道念珠菌病。Objective To investigate the effects of Berberine and Matrine, both of which are active compo- nents from Chinese materia medica, against the formation of mixed bacteria biofilms of vulvovaginal candi- diasis in vitro. Methods Mixed biofilms of Monilia albicans and Pseudornonas aeruginosa were developed in vitro. Tetrazolium salt (XTT) reduction assay and morphological methods were employed to observe the formation process of mixed Monilia albicans and Pseudornonas aeruginosa. The sessile minimal inhibi- tory concentration (SMIC) of Berberine and Matrine on mixed biofilms of Monilia albicans was evaluated using morphological methods and colony-forming unit (CFU) counting. Results Monilia albicans formed mature biofilms after being cultured for 48 hours. Berberine 125 mg/L could inhibit the formation of mixed Monilia albicans. Matrine 500 mg/L was not found to inhibit the formation of mixed Monilia albicans. Conclusion Berberine can be used for preventing vulvovaginal candidiasis because of its inhibitive effects against the formation of biofilms.
