猪2型圆环病毒新疆株全基因组的克隆与序列分析

摘　　要：根据GenBank中猪2型圆环病毒(PCV2)的核苷酸序列,设计了一对引物,采用PCR方法从疑似断奶仔猪多系统衰竭综合征(PMWS)的死亡仔猪组织病料中扩增出一株PCV2全基因组,将其克隆到pMD simple18-T载体上,筛选获得了重组质粒pMD18-T-PCV2。对此重组质粒进行序列测定及同源性分析,结果表明,该PCV2新疆株的全基因组大小为1 767bp,与其它PCV2核苷酸序列同源性为95.0%～99.6%,而与PCV1的同源性仅为76.8%～76.9%。A pair of primers was designed according to the published complete genome of PCV2,and the complete genome was amplified from the suspected PMWS sample by PCR.The PCR product was cloned into pMD simple18-T vector,and the recombinant plasmid pMD simple18-T-PCV2 was obtained.The sequence of the cloned PCR product was analyzed and compared with other PCV2 isolates in GenBank.The results indicated that the complete genome of PCV2 Xinjiang isolates was 1 767 bp,it shared 95.0%~99.6% nucleotide homology with other PCV2 isolates in GenBank and 76.8%~76.9% nucleotide homology with two PCV1 isolates.
