摘 要:目的探讨人血清白蛋白(HSA)和依他尼酸的相互作用。方法通过荧光光谱、紫外可见吸收光谱、傅里叶变换红外光谱、圆二色谱、分子模拟技术考察两者的相互作用。结果利用Scatchard方程确定了两者相互作用的结合位点数以及结合常数;根据Van't Hoff方程求得了相互作用的摩尔焓变、摩尔熵变和吉布斯自由能变;计算出了两者相互作用的距离;利用分子模拟技术证明了两者之间主要的作用力是疏水作用力和氢键。结论 HSA和依他尼酸间存在相互作用,可改变HSA的二级结构。Objective To study the interaction between human serum albumin(HSA) and ethacrynic acid.Methods Fluorescence spectra,UV-Vis,FT-IR,circular dichroism spectroscopy and molecular modeling were used to study the interaction between HSA and ethacrynic acid.Results The number of binding sites and binding constants were determined by using the scatchard equation;the enthalpy changes,the entropy changes and gibbs free energy changes were calculated according to the Van't Hoff equation.The distance r between ethacrynic acid and HSA was calculated.Molecular modeling revealed that main interaction between ethacrynic acid and HSA was hydrophobic forces and hydrogen bonding.Conclusion There is interaction between HSA and ethacrynic acid,and this effect can change the secondary structure of HSA.
