银黄片中绿原酸和黄芩苷多波长薄层扫描测定

摘  要:采用多波长薄层扫描法同时测定银黄片中绿原酸和黄芩苷的含量,将阵列感光的光纤光谱仪与二维电移平台结合,采取"弓"形二维移动扫描,探头与薄层板间距2 mm,波长范围190 nm~1 100 nm;以质量分数36%乙酸为展开剂在聚酰胺薄膜上展开.结果表明,绿原酸和黄芩苷对照品溶液点样量分别在0.22μg~0.77μg,0.4μg~1.40μg范围内吸光度积分值均具有良好的线性关系,r分别为0.996 8,0.997 5;平均回收率(n=6)为分别为98.64%,96.23%,RSD分别为2.49%,0.97%.该方法扫描定量精确度较高,操作简单,可以应用于银黄片中绿原酸和黄芩苷的含量测定.This papper aims to determine the content of chlorogenic acid and baicalin by Multi-wavelength TLC scanner.By combining spectrometer of array sensor with 2D moving stage to scan and record the multi-wavelength reflectance of thin-layer plate,the scanning track was designed in shape of "S",and distance between probe and plate was 2mm,the range of detection wavelength was 110-1900nm;The separation was achieved on polyamide film with 36% ethanoic acid as mobile phase.Calibration plots of chlorogenic acid and baicalin were linear in the range 0.11-0.77mg.mL-1 and 0.40-1.40mg.mL-1.Fiber-optic spectrometer scanning showed that the correlation coefficient of the two components respectively were 0.9968,0.9975.The recoveries of chlorogenic acid and baicalin were 98.66% and 96.23%,and the RSDs respectively were 2.49% and 0.97%.With the characteristics of high scanning quantitative precision,simple operation,this densitometric method can be used for quantitative analysis of chlorogenic acid and baicalin in YinHuangPian.
