摘 要:SSR标记技术是农作物品种纯度鉴定主要手段之一。本研究介绍了一种适合油菜杂交种纯度鉴定的快速SSR检测方法,检测时间比目前常规的SSR检测缩短5h,且操作过程不需要研磨、离心沉淀、冷冻、晾干和溶解等操作,每人每天可完成480个单株的检测。将该方法用于2个油菜杂交品种制种样品的纯度检测,并与醇溶蛋白电泳和田间种植的鉴定结果进行比较,结果表明醇溶蛋白电泳和快速SSR检测结果差异不显著,且极显著相关(r=0.991),田间种植鉴定结果高于上述两种方法,但相关性均达到显著水平。该检测方法可以为油菜杂交种纯度鉴定提供可靠依据。SSR marker based technology is one of major approaches for identifying crop variety purity. In this research we developed a rapid SSR based detecting method, hereinafter called the SSR based method, that wassuitable for purity identification in rapeseed varieties. Comparing with the existed SSR based methods, the procedures of this method simplified some complicated steps leading to save detection time 5 hours, bypassingmechanical grinding, centrifuging, refiigerating, drying and dissolving. Detecting 480 samples can be easy completed by a person a day. Comparative analysis of the SSR based method with the alcohol-soluble proteinPAGE method and field trial test was carried out to identify the purity of two hybrid rape seed varieties, the results showed that there was no significant difference between the SSR based method and alcohol-soluble protein PAGEmethod with significantly correlation (r=0.991) in statistics, whereas the values of purify of the field trail test were generally higher than that of the other two methods with the significantly correlation. This method can provide thebasis for purity identification of hybrid in Rapeseed.
